Alamar Blue Assay Lab Report
Alamar Blue assay are performed to show cell viability and proliferation. In the procedure we treated our cell line, SUM 149 cells with different concentrations of an unknown treatment. SUM 149 cells are an inflammatory breast cancer cell line. The treatments were used to suppress cell growth and proliferation. The station that I performed my work was station C. I platted my cells and aloud a previously determined allotted time for cell proliferation and growth to occur. The cells were then treated with varying concentrations of an unknown treatment to be administrated. Once the treatment was performed an Alamar Blue assay was then ran to assesse the effects of the treatment. The active ingredient in Alamar Blue is resazurin, which is reduced to resorufin that is red and fluorescent. The …show more content…
The solutions 1-4 have readings closer to 0 than 1000. My control contained cells plus medium that were not treated has a reading approximately around 6,700. This contrast shows that the cells in the different concentrations of unknown treatment had low viability. The cells+medium control has high viability. A higher fluorescence reading means that there is a higher cell level. Solution two’s concentration was the most effective if the purpose of the unknown solutions is to hinder cell viability. The trend that the treatment solutions followed was dose dependent. This is due to the fact that as the concentration changed the viability changed the viability changed in a direct manner. This could not be considered threshold concentration due to the fact that the desired result was to limit cell viability and all of the cells that were treated showed a desired affect. Solution five is just another concentration and not a vehicle control because if it was then the vehicle control should have the same reading or close to the same reading as the cells+medium

    Natural enzymes are proteins that catalyze biological reactions by lowering the activation energy of the reaction without being altered during the process. The enzyme used in this experiment was the β-galactosidase purified from E. coli. This enzyme hydrolyzes lactose and turns it into galactose and glucose. Since it is difficult to assay the activity of β-galactosidase, we will be using the artificial substrate, o-nitrophenyl-β-galactoside (ONPG) instead of lactose. ONPG is an analog of lactose and an advantage of using ONPG is that it is easy to determine the amount of ONPG cleaved by using spectrometric assay (1). The β-galactosidase hydrolyzes ONPG and yields a yellow solution that contains o-nitrophenol and galactose. The solution becomes more yellow as the more ONPG is being degraded. Using spectrophotometry, the absorbance of the solution can be determined at a wavelength of 420nm. The assays will help determine the Km, Vmax, and Kcat of the enzyme.     The first plate was labeled with our initials, lab section, one half was called the exposed and the other half was called foiled protected. The second had four sections labeled- Coconut oil, SPF 50, Sunglass and Control. Next, 0.15 ml of yeast was pipetted onto the two separate agars. Then the disposable L-rod spreader was used to spread the liquid so that the agar surfaces are evenly covered. Then the plates were placed in a dark place for 20 minutes.
