Abstract
Experiment 5 & 6 are conducted to familiarise the students with the use of spectrophotometer to study the effect of temperature on the enzyme and to study and measure the enzyme activity using spectrophotometer respectively. For Experiment 5, a mixed solution of ONPG solution and Sodium phosphate buffer pH 7.3 is incubate at different temperature, 25, 40, 50, 60 and 70 °C. The mixed solution is then incubated for another 10 minutes after the enzyme is added in their respective temperature.
