A gram-negative bacteria like Escherichia coli protect its cells from stresses that cause the misfolding of proteins, which are later secreted across the cytoplasmic membrane (Raivio et al., 2013). In response to the secretion of misfolded proteins, gram-negative bacteria use two major envelope stress responses (Raivio et al., 2013). One of the envelope stress responses is the Cpx envelope stress response and it is activated by a diverse set of inducing signals, such as alkaline pH, chloride ions, copper, mutations that affect the folding of proteins in the periplasm, the overexpression of misfolded and aggregated periplasmic proteins at the inner membrane, and attachment to abiotic surfaces (Raivio et al., 2013).     The purpose of this experiment was to show the genetic transformation of E. coli bacteria with a plasmid that codes for Green Fluorescent Protein (GFP) and contains a gene regulatory system that confers ampicillin resistance. A plasmid is a genetic structure in a cell that can replicate independently of chromosomes. In this lab, the Green Fluorescent Protein, which is typically found in the bioluminescent jellyfish Aequorea Victoria, was cloned, purified, and moved from one organism to another with the use of pGlo plasmids. It was hypothesized that if bacteria that were transformed with +pGlo plasmids are given the gene for GFP, then transformed cell colonies will be located on the LB/amp/ara and LB/amp agar plates. Cells that have been transformed with +pGlo plasmids have the ability to grow in ampicillin plates, and the arabinose sugar allows the colonies to be visibly fluorescent under ultraviolet light. The GFP is able to resist ampicillin because of the Beta- Lactamase protein that is produced and secreted by the bacteria that have been transformed to include it in their plasmids. Arabinose is a carbohydrate, normally used as a source of food by bacteria. Bacterial colonies are not able to grow on –pGlo plates because they are sensitive to ampicillin. We used heat shock in order to make the bacteria capable to uptake a plasmid in the presence of calcium ions that help disrupt the cell membrane (heat shock is the combination of altering hot and cold). However, the need of an ion gradient to power ADP phosphorylation is central to all ATPases. Here, we focus on bacterial cell membranes, particularly E. coli, as they possess a unique Mg2+/Ca2+ ATPase. Several ATPase assays containing Pi, Mg2+ or no Mg2+, and DCCD were conducted where the reverse reaction -ATP hydrolysis into ADP and Pi- is used as an approximate measure of ATPase activity and dependence upon Mg2+, as well as the effects of inhibition by DCCD on Pi production. The ion gradient needed to power this ATPase will also be of importance as we determine what substrates the ETC preferentially oxidizes to produce the necessary H+ ion gradient.
