Research Question: E. coli strain MM294 is placed under different sound frequencies. Does sound frequency affect the growth of E. coli strain MM294?
Background:
Noise pollution is a problem in urban areas and can be the cause of several health issues and increase risks of disease. It is interesting that even something inanimate like sound could affect so much of biological processes. The movement of energy through matter produces sound. Sound is a mechanical wave that travels in waves and has the ability to move particles. Noise pollution is the result of the rapid and loud buzzing of a city, factory, piece of machinery, etc. The vibrations caused by energy vibrate the air particles around it and produce sound waves. I made the sound load boxes by adhering the pads to the panels then placing the panels together in a cube and adhering them with glue and nails but left the top unsecured. I constructed the noise cancelling compartment out by cutting the padding in order to hold a petri dish. I cut three whole in the padding and layered four pads to leave the empty space in the middle. I placed the plywood panels on the top and bottom and secured the bottom and left the top unsecured for access. I made 2 liters of agar and poured it evenly into 12 petridishes with an area of 18.096 cm2. Each Petri dish was inoculated with an inoculation tube using sanitation methods and intending to keep the amount of bacteria added constant. The bacteria was then set into the sound load chamber. The speakers were turned on according to the frequency being tested then the dab level was set for the sake of being consistent. The bacteria was then observed after 48 hours and the plate coverage was recorded by measuring the bacterial colonies and calculating their area respective to the total are of the plate.     In this experiment we will inoculate individual vials of nutrient broth with various organisms that are known to be resistant to multiple antibiotics. We will also inoculate various organisms that are as close to the wild type as is feasible. The vials will be allowed to incubate at 37 degrees Celsius for 24 hours. The number of bacteria in a given cultured sample is too great to be counted directly. The samples will be serially diluted by a factor of 10 in sterile saline, to be repeated 7 times. A 10 microliter aliquot from each dilution will be placed on a 5% blood agar plate or a MacConkey plate, depending on the type of organisms plated. These plates are selected because of the ease of differentiation between organisms. The original vial and all diluted aliquots will then be refrigerated to restrict growth until the plates can be processed and evaluated. The plates will be streaked in the standard format for colony count. The plates will be incubated at 37 degrees Celsius for 24 hours. The plates will then be examined for purity of culture and for colony count. The number of colonies will be counted for each dilution.
