Effect of Catalase at Different Temperatures
Effect of Catalase at Different Temperatures
Abstract
The role of this experiment is to determine the effect of temperature on enzymatic activity. The results of the experiment were the colder the temperature the slower the reaction rate and the hotter the temperature the faster the reaction rate.

Introduction
Enzymes are chemical substances found in living cells and they act as catalysts of the various chemical reactions that occur in them.(Preszler, 2012) They bind to substrates that have binding sites that are specific to each enzyme and remain unchanged by the reaction. Enzymes work best at a certain temperatures. Too cold, and it's very sluggish. Too hot, and the enzyme breaks down (denatures). Therefore the hypothesis of the experiment is that the colder the temperature becomes the slower the reaction rate.
This hypothesis was tested by placing enzymes solution (catalase) in three different temperatures, the colder the temperature the slower the enzyme soaked paper would take to reach the top.

Methods
To find the effect of temperature on the activity of an enzyme, the experiment deals with the steps as follows. First, 3 beakers filled with the enzyme solution, 3 test tubes with the hydrogen peroxide, 3 pieces of the filter paper. These three test tubes were set at three different temperature settings. The first test tube was placed in dry ice for three minutes until it reached a temperature of 0° C or less. The second tube’s temperature setting was at room temperature until a temperature of 22°C was reached. The third tube was placed in a warm-water bath until the contents of the beaker reached a temperature setting of 37° C. The beakers were also put in the three different temperatures. Once both were out the filter paper was soaked into the enzyme solution. The paper was dropped in the hydrogen peroxide, once the filter paper hit the bottom a stop watch was use to time the paper as it floated up to the top. This was done 6 times total. The independent variable is the temperature. Dependent was the time the paper took to float to the top.
Results

The reaction times are all different with each temperature. The test tubes in the warm water bath had a faster reaction rate, room temperature was a medium rate and dry ice had the slowest reaction rate.

Discussion
This experiment on enzymes was done to help convey how there are environmental factors that have an effect on the activity of an enzyme. From the results that were given the hypothesis was in fact true. The colder the temperature becomes the slower the reaction rate. The warm bath water was the faster of the two. Room temperature was in the middle, with not to fast nor slow. In conclusion I think the experiment on enzymes was an interesting one and most important the hypothesis was correct.

The prediction for the effects of temperature on the enzyme activity was that the reaction’s rate would increase as the temperature increased, until they go over the optimum temperature where the enzymes denature and the reaction’s rate quickly drops to zero. At 5 degree C the rate is 0.00059mole PNP/min. This then increases to 0.01031mmoles PNP/min at a temperature of 50 degree C. The rate then drops drastically to -0.00215moles PNP/min. This point is where the enzymes have been denatured and have no activity, shown as the last point on the fig 8 and 9, do not fit on the graph. The optimum temperature was about 47 degree C. The core body temperature is only about 37 degree C and thus these enzymes are operating below their optimum temperature.…

If the temperature was increased, the rate of reaction slows. If the temperature of the HCl was raised by 10°C, the activity will increase by 50-100%. This did not happen with the results shown in the chart above. Even small temperature changes such as 1-2°C can change the effect of the enzymes by 10-20%.…

Introduction: The Enzyme Lab is to conduct investigations to determine the most favorable conditions for the most efficient enzyme activity. Variables to be used testing include temperature, pH values and surface area. Enzymes are proteins that speed up the rate of chemical reactions, which would otherwise progress more slowly.(Background Information; pg. 1) pH is a measurement of the acidity or alkalinity (base) of a solution. When the liver got mixed with H2O2 , the first time the chemical reaction was fast, the second time the reaction was slow and the last try was very fast. Temperature is the degree or intensity of heat present in a substance or object. When the temperature of the liver changed from freezing to very hot to room…

Enzymes are organic catalysts that spur metabolic reactions. The presence of an enzyme within a cell is essential in order for any sort of reaction to take place. All enzymes are complex proteins that act in an organism's closely controlled internal environment. In such a homeostatic environment, the temperature and the pH (concentration of hydrogen ions), remain within a fairly narrow range. Extreme variations in pH and temperature denature the enzyme by altering its chemical structure, thus adversely affecting the chemical reaction. As even the slightest change in the protein's structure will change the enzyme's shape enough to prevent the formation of the enzyme-substrate complex, the reaction will then be unable to take place. The enzyme, catalase, reacts with hydrogen peroxide in order to yield water and oxygen. Therefore, upon reaction, bubbling will occur as oxygen is produced. The amount of bubbling, or oxygen that is produced is dependent on the conditions under which the reaction takes place (or fails to).…

In our everyday lives, enzymes are used in our bodies, and in nature around us, to speed up the chemical reactions happening constantly, which happens by lowering the amount of activation energy needed to start various reactions. The way this works is by attaching the particular substrate to the active site of the enzyme, where it will start to aid the chemical reaction. Then, the allosteric site involves itself in forming the final 3D shape. For each specific reaction is a specific enzyme that helps speed up the reaction, and the reason for the variations of the enzymes is their unique protein structures. However, this means that once the structure of the enzyme is denatured and changed, the functions will most probably modify as well. In nature, this happens when the temperature and concentrations of different components are altered. In this lab experiment, we will be doing an in-depth research of exactly what happens to the enzymes, when it happens, and why it denatures the way it does.…

Introduction: Enzymes are biological catalysts that speed up metabolic reactions without being affected. They lower the activation energy needed to start a reaction.Enzymes are affected by several factors including PH, Substrate concentration; Temperature & other factors. Each enzyme has an optimum temperature at which its activity is the highest, below this optimum temp, the kinetic energy of molecules decrease , therefore the collisions between the active site of the enzyme and substrate decreases , as a result the enzyme activity will decrease , so decreasing the rate of the reaction If the temp. Exceeds the optimum temp. The kinetic energy between molecules increase therefore collisions increase leading to the change in the tertiary structure of the enzyme and in this case active site is lost and the enzymes will be denatured so the reaction will slow down &stops. Catalase is an enzyme, found basically in all living cells. It breaks down hydrogen peroxide (waste product) into water and oxygen.…
