Faiyaz L Investigate Factors Affecting Cell Membrane Permeability In Beetroot
Investigate factors affecting cell membrane permeability in beetroot.
Research Question: How does variation in temperature affect the permeability of cell membranes?
Background Information: In this investigation I will want to identify how variations in temperature effect the permeability through the use of a beetroot. I predict that as the temperature of the water increases so will the dispersion of the pigment.
Independent Variable: The temperature of water will be increased in increments of 10oC, thus the temperatures I will be using are 20oC, 30oC, 40oC, 50oC and 60oC.
Dependent Variable: I will be measuring the cell membrane permeability through the amount of pigment dispersion. A colorimeter will be used to quantify results into a percentage transmission of light, after it is left for 5 minutes.
Control Variables:
Variable
How it is measured
Why it needs to be measured
Mass of beetroot cylinder
A weighing scale should be used to ensure all beetroot cylinders weigh 1 gram.
If the mass of the beetroot cylinder varies it may cause a difference in diffusion capabilities.
Length of beetroot cylinder
The in order to ensure the surface area is kept the same, each cylinder must be 2 cm long. This should be measured using a 15cm ruler.
If the lengths are different it would result in a difference of surface areas therefore questioning the test’s accuracy.
Age of the beetroot
To keep the age of the beetroot the same, the same beetroot will be used to extract the cylinders.
Different ages can cause the amount of pigment in the beetroot to fluctuate. Therefore it is vital to use the same beetroot.
Volume of water
A 25ml measuring cylinder should be used to accurately measure 15ml of water for each test tube.
Water is a solvent and only a certain amount of pigment can be absorbed by the water. Hence it would create a contrast in water potential.
Time
Each cylinder will remain in its test tube for 5 minutes before the water is removed and tested. A stopwatch must be used.

    Betalin - Beetroot

    • 1357 Words

In cooked beetroot, the water or food would become stained due to its leakage of betalin (the redish-purple pigment). Betalin is found in the vacuole of beetroot cell. The leakage may have been caused by something happening in the cell membrane of the beetroot – concerning the proteins embedded in the phospholipid layer. The study will require the experiment of different temperatures of water and to find out which in temperature causes the most leakage of the pigment and why it does this.…

When a beetroot cell is exposed to various kinds of temperatures, its plasma membrane is affected and may change in structure resulting in the leaking of betacyanin. If the temperature changes, then I would expect to observe that there would be a change in the concentration of betacyanin that has leaked from a beetroot cell. In specific, at high temperatures, the phospholipids that make up the plasma membrane of the beet root cell become progressively more destabilized causing them to transform into a liquid state. As this occurs, the plasma membrane ruptures allowing the betacyanin pigment to leak out. This is why we would expect to see a higher concentration of betacyanin leakage at higher temperatures. As the results of this lab indicate, as temperature increases, the plasma membranes of beet root cells (that were immersed in high temperatures of water) became damaged, allowing for more betacyanin to leak out resulting in a higher concentration of the pigment. What was unexpected, however, was that at -5ºC, the amount of betacyanin that leaked from the beetroot cell was higher than any of the other temperatures that were used to measure betacyanin concentration. This result does not support my hypothesis. But, it illustrates how temperatures that are far from ideal growing conditions for beetroot (around 15ºC to 19ºC) may result in a larger leakage of betacyanin from the beetroot (Nottingham 2004). The rest of the results support my original hypothesis, which explained how high temperatures affect the form of the phospholipids, which in turn change the structure of the plasma membrane. But, the entire set of results support a new idea that the farther that temperature strays from ideal beet root temperatures, the more damage is done to the membrane of a beet root cell, which in turn causes more betacyanin leakage.…
