4.2.3.1 C) HPTLC Phenol profile of M. concanensis Nimmo.

HPTLC finger printing of M. concanensis was done by using selected solvent system Chloroform: Ethyl acetate: Formic acid (50%:40%:10%v/v) for leaf, flower and seed extracts, visualized under UV 254 and 366 nm showed more components. The Rf values and chromatogram in the methanol extract of leaf, flower and the seed was presented in the Table-8. In general, more degree of phyto compounds diversity has been observed in vegetative part when compared with the vegetative parts. The methanol extract of leaves, flowers and seeds of M. concanensis showed the presence of 33 peaks with different types of Rf values with range from 0.01 to 1.04. A total number of 6 peaks with Rf values 0.01, 0.41, 0.47, 0.73, 0.84 and 1.04 for leaves, 13 peaks with Rf values 0.11, 0.26, 0.40, 0.49, 0.54, 0.69, 0.71, 0.78, 0.84, 0.92, 0.96, 0.98 and 1.02 for flower and 14 peaks with Rf value 0.03, 0.26, 0.35, 0.40, 0.53, 0.55, 0.62, 0.71, 0.77, 0.85, 0.93, 0.95, 1.00 and 1.03 were observed in seeds. The maximum phenol content of M. concanensis was observed in leaves followed by flowers and seeds. This is in harmony with phenols standard of Gallic acid, the Rf value being 0.85 present in the seed extract showed in the plate-19.

    An experiment was conducted to determine the effect of the allelopathic compounds of Encelia Farinosa (brittle bush) on desert native annuals and non-native seed germination. The experiment was set up by putting native wild flower seeds in six Petri dishes and lettuce seeds in six different dishes. To three of each dish containing the different seeds, water was added as a control. To the three other dishes, a prepared brittle bush tea (filtered ground brittle bush leaves and water) was added. After a week’s time, the number of germinated seeds in each dish was counted. The results showed that the allelopathic compound in brittle bush did have a significant effect on both the lettuce seeds and the native wildflowers when compared to their respective controls. However, the results showed no significant difference between the two means of the native seeds with brittle bush tea and the lettuce with brittle bush tea.     The chromatography of the spinach leaves was taken to establish what pigments were present in the chloroplasts of the leaves. The solvent carried and separated the different pigments up the cellulose paper. The solvent moved up the paper because of capillary action because of adhesion of the solvent molecules and the paper. The pigments dissolved in the solvent also were carried up the paper and were separated because they do not all dissolve equally into the solvent, less soluble pigments appear lower on the paper. The pigments also were attracted to the paper at different levels. Each pigment has a specific Rf value which is the distance the pigment migrated divided by the distance the solvent migrates. The data shows that Chlorophyll a has the highest Rf value, and it did travel the farthest out of the pigments. The control in this experiment was the same spinach leaves used. Error was no Carotene detected. Carotene is the very abundant in plants, so the lack of it found was based on reading errors of the chromatography, or the confusion of Xanthophyll with Carotene because the Rf value of Carotene is 0.98, which was calculated for Xanthophyll.…

    The TLC plate showed there are total four components in spinach. The Rf value of the first spot, second spot, and third spot are 0.5882, 0.9059, and 0.9059 respectively.
