HPLC
High-performance liquid chromatography (formerly referred to as high-pressure liquid chromatography), HPLC, is a chromatographic technique used to separate the components in a mixture, to identify each component, and to quantify each component. HPLC is considered an instrumental technique of analytical chemistry (as opposed to a gravimetric technique). In general, the method involves a liquid sample being passed over a solid adsorbent material packed into a column using a flow of liquid solvent. Each analyte in the sample interacts slightly differently with the adsorbent material, thus retarding the flow of the analytes. If the interaction is weak, the analytes flow off the column in a short amount of time, and if the interaction is strong, then the elution time is long. HPLC has been used in medical (e.g. detecting vitamin D levels in blood serum), legal (e.g. detecting performance enhancement drugs in urine), research (e.g. separating the components of a complex biological sample, or of similar synthetic chemicals from each other), and manufacturing (e.g. during the production process of pharmaceutical and biological products).[1]
Chromatography can be described as a mass transfer process involving adsorption. HPLC relies on pumps to pass a pressurized liquid and a sample mixture through a column filled with a sorbent, leading to the separation of the sample components. The active component of the column, the sorbent, is typically a granular material made of solid particles (e.g.silica, polymers, etc.), 2–50 micrometers in size. The components of the sample mixture are separated from each other due to their different degrees of interaction with the sorbent particles. The pressurized liquid is typically a mixture of solvents (e.g. water, acetonitrile and/or methanol) and is referred to as a "mobile phase".     Gas Chromatography separates organic samples much in the same way as column chromatography. The only differences are that it uses a moving gas phase and a stationary liquid phase, and that the temperature of the gas system can be controlled. In a gas chromatograph the sample is shot in with a syringe and is immediately vaporized in a heated injection chamber. It is then introduced to a moving stream of gas called the carrier gas which sweeps the vaporized sample into a column filled with particles filled with liquid adsorbent. This column is usually filled with liquid that has a low vapor pressure and high boiling and is called the stationary phase. This phase is also usually coated onto a support material very evenly and packed into a tubing apparatus as evenly as possible and placed in the temperature controlled oven. When organic solutions are passed through the tubing van der Waals forces attract the nonpolar molecules especially if they have large molecular weights. Polar molecules can be attracted in many ways. Interactions include salt formation, coordination, hydrogen bonding, and even dipole-dipole. Through these interactions the molecules in the vaporized sample will separate accordingly.     Column chromatography was invented by Tswett in 1906 so that his study of the chemistry of chlorophyll could be facilitated. Tswett made significant advances in the method and demonstrated its applicability to the separation of many different kinds of mixtures. Column chromatography rapidly found application in all areas of chemistry and is used extensively today because it is a simple but powerful tool for the separation of pure compounds from a complex mixture.     Chromatography is a term used to define a set of laboratory techniques for the separation of mixtures. Through Chromatography we analyzed complex mixtures. In our recent lab we analyzed complex mixtures. The purpose of this lab was to find whether the water soluble or acetone soluble liquid traveled farther. In our recent lab we measured the distance traveled by the different colored water and acetone soluble inks. W put three dots of water soluble ink on a piece of filter paper and four dots of acetone soluble ink at the other end. We then put the end of the filter paper in a small amount of distilled water in a beaker. The filter paper absorbed the water and the ink slowly spread to the top of the paper. We repeated these steps for both the water soluble and acetone soluble ink but when we did the acetone soluble ink, we placed the filter paper in an acetone-water mixture instead of just distilled water. We then recorded our results which showed that water soluble inks traveled farther than the acetone soluble inks. The results of our experiment showed how each different color of water soluble inks traveled farther than any of the acetone soluble inks.…

    Extraction is one way of purifying a substance. In an extraction, a solute is transferred from one solvent to another. Extractions depend on polarity and solubility; polarity is the separation of charge caused by a difference in electronegativity, and solubility is the ability of a material to dissolve into another physical phase.
