HPLC Analysis of Caffeine
Date: 9-10-13
Title: HPLC analysis of Caffeine
Lab Partners:
Objectives:
1) The main objective of this lab is to gain experience in using and reading results from the HPLC machine.
2) The first step is to inject a series of caffeine standards into the machine in order to get results/values of peak height and area.
3) The next step is to investigate the effects of a series of HPLC parameters on Retention time, peak height and peak area.
4) To use above data from peak height and area to construct two standard curves vs concentration.
Introduction:
Caffeine is a common organic molecule found in many beverages such as coffee, tea, and cola. It is a stimulant to the central nervous system. Which is why the majority of the population use it in one form or another to help stay alert. In general people drink caffeinated drinks like the ones mentioned above in order to obtain their “fix”. However with many recent studies in the area caffeine tablets are becoming a very popular way for athletes in lots of different sports to stay alert even when fatigue is setting in. HPLC stands for High Performance Liquid Chromatography but is also referred to as High Pressure Liquid Chromatography. It is used for separating mixtures either to analyse the mixture or to separate a required product from others in a reaction mixture. It can also be used to find the relative amounts of different components in a mixture. HPLC works along the same lines as paper chromatography. In paper chromatography a liquid (mobile phase) moves past a solid (stationary phase). In paper chromatography the stationary phase consists of water molecules bound to the cellulose in the paper, the mobile phase carries different components of the mixture along with it. How fast each one moves depends on its relative affinity for the mobile and stationary phases. In HPLC the stationary phase is a solid packed into a column and the liquid is forced through the column by high pressure pumps.

After the 10ml of dichloromethane was added to the separatory funnel, two layers began to form. The top layer is the organic layer and contains the caffeine while the aqueous is at the bottom. The dichloromethane has a density of 1.326g/cm so it is denser than water and thus is at the bottom of the funnel. After the distillation , the Bunsen burner was used to sublimate the caffeine as the heating mantle and the water bath would take to long for it sublimate. The text book “Small scale approach to organic laboratory techniques” on pg.77 dictates that “Caffeine constitutes as much as 5% of the leaf material in tea plants.” Using that percentage, this estimates the amount of caffeine in 5 grams of tea leaves is 0.25 grams compared to the actual 0.017grams gained and 6.8% was collected from the 5 grams. This lab performance was better than the acetaminophen lab and overall cooperation of participants was surprising though one of the participants confused the reflux apparatus with the…

Abstract: Caffeine, (C8 H10 N4 O2) a white, crystalline alkaloid known as a nervous system stimulant found commonly in tea leaves and coffee, can be extracted using the method of sublimation with the use of Methylene Chloride (CH2Cl2) and Sodium Hydroxide; both can repeatedly rinse the green (unroasted) coffee beans from the caffeine.…

When a person drinks tea or coffee, it is not pure caffeine. It also has other natural substances. The purpose of this lab was to isolate caffeine from tea leaves. The methylene chloride removed nearly pure caffeine from the basic tea solution. Caffeine is a white, crystalline, bitter alkaloid, C8H10N4O2 , usually derived from coffee or tea. Caffeine is used in medicine chiefly as a nervous system stimulant. Caffeine is used by humans to ward off drowsiness and restore alertness. Caffeine constitutes as much as 5% by weight of the leaf material in plants. The hypothesis for this experiment is that the observed melting point of the pure caffeine will be lower than the melting point of pure…
