Kola Nut Experiment
This work is focused on evaluation of the potential anticancer effect of a kola nut extract on the human immortalized myelogenous leukemia line (K562) and the human immortalized line of T-lymphocyte cells (Jurkat). The objectives of this research are as follows:
1- Does kola nut extract inhibit leukemia?
2- Does the growth in inhibition induced by kola nut extract fellow a dose- and time- dependent response?
3- Does kola nut extract induce apoptosis or necrosis?
4- Which cell line, a K562 or Jurkat cell line is more sensitive to this targeted extract?
4-METHODOLOGY
4-1 Study design
The design of our study is a type of case-control design, where the induction of apoptosis on both the myeloid (K562) and lymphoid (Jurkat) human cell lines by kola nut extract will be investigated. Moreover, the cells will be divided and sub-cultured into T-75 culture flasks once a week at ratio of 1:5. The targeted action of apoptosis induction by Kola nut extract will be investigated in both cell lines by several screenings and specific methods as following below.
4-5 Kola Nuts Extraction:
Kola nuts will be extracted in a standard method as previously performed in many published works. Several solvents including water, acetone, ethanol and methanol will be used in our extraction procedure to determine the proper solvent with acceptable toxicity effect over cells and maximum-targeted extraction. The proper concentration and incubation time with the targeted extract will be optimized through a pilot study.
