Lab2: Testing for Proteins and Starches

In this lab 8 total substances were tested to find out whether they are a Protein or a Starch.

It is my belief that only 1 or two of each of the substances in test 1( proteins) and test 2 (starch) will test positive for either protein or starch.

For this lab the following materials were needed to complete the experiments in test 1 for proteins: Di water, ev milk, 50% egg solution, 1% sucrose, 4 test tubes, 1 test tube rate, safety glasses, pipets and the reagent solution. For test 2 for starches the following materials were used: Di water, 1% starch solution, onion juice, sucrose solution, iodine reagent, 4 test tubes, test tube rack and pipets.

In test 1 and test 2 each solution was placed in a separate test tube with the corresponding reagent added to each as well. In test one for proteins each substance that showed a purple color it indicated a positive result for proteins. Any solution that stayed blue is to be considered negative for proteins. The Di water in test tube one resulted in a blue color once the reagent was added. In test tube 2 Evaporated milk had the reagent added for a resulting color of milky purple. In test tube 3 the egg solution the color turned purple once the reagent was added. Finally for test one in test tube 4 containing sucrose turned light blue once the reagent was added for a negative result.

In test 2 for starch the first solution used was Di water as the control similar to test 1. Test tube one resulted in a yellow color and a negative test. Test tube 2 with 1% starch turned light purple once the iodine reagent was added. The iodine reagent is used in test two because it is the only reagent that will indicate traces of starch. In test tube 3 onion juice turned yellow once the iodine was added and finally in test tube 4 the sucrose solution also turned yellow.

For both tests 1 and 2 the results were as expected. In test 1 for proteins the solutions evaporated milk and 50% egg both

    We then repeated this in two more test tubes. We put one of the test tubes in ice (0 C) we put another in the water bath (42 C) then we put one in boiling water (100 C). We repeated the same thing for the boiled amylase. We then checked at five minutes to see if the starch had been digested by the amylase. We did this by putting a little of each of the six samples into a spotting tile. We then added iodine solution to this to see if starch was present or not. I could tell if starch was present or not because if the solution went orange it meant that there was no starch present, so all the starch had been digested, if the solution went blue/black it meant that there was starch present, so the starch had not been fully digested.     In my first experiment to test for proteins the egg white with the Bluret reagent should come out positive for being a protein. The pepsin in the Bluret reagent should be positive and should be a protein. The test for sugar in the Bluret reagent should be negative and I don't think it is a protein. The test with the distilled water with the Bluret reagent should be negative and is not a protein. In the second experiment to test for glucose with the Benedict reagent sugar should be positive for glucose. Starch mixed with the Benedict reagent is probably positive for glucose because it is a polysaccharide. Onion juice mixed with Benedict reagent is probably positive for glucose because it is a fructose. Distilled water with Benedict reagent should be negative for glucose. The third experiment to test for starch using with and without the IKI reagent by looking under a microscope a slice of potato cells should have several shapes to it and with the IKI reagent should look have more of a stronger shapes and color to it.     These biological macromolecules are to be identified by the changes in colour through three different tests - Iodine Test for starch and glycogen, Benedict 's Test for reducing sugars, and Biuret Test for Proteins. However, only two macromolecules are being identified in this experiment - carbohydrates and proteins. There are 12 solutions to be tested in this experiment. The Iodine test is used to indentify starch and glycogen in the given solutions. Of the 12 solutions, solution 8 is a starch solution and solution 7 is a glycogen solution. Starch solutions turn blue-black when Iodine solution is added to it. This is due to the formation of polyiodide chains when the Iodine solution mixes with starch. Starch contains both amylose and amylopectin. The amylose molecules in starch form helices at the locations where the Iodine molecules assemble. This cause a dark blue-black colour change ("Starch-iodine test", 2008). Therefore, solution 8 should turn blue-black when Iodine solution is added to it since it is a starch solution. However, glycogen solutions turn red-brown when Iodine solution is added. The chemical structure of glycogen is similar to the structure of amylopectin. Glycogen is highly branched.
