TITLE: EXTRACTION OF DNA

OBJECTIVE:
Extract DNA from yeast, onion and apple.

INTRODUCTION:
Every cells of living organism contains genetic materials known as deoxyribonucleic acid, DNA. It can be isolated from tissue sample of living things by separating it from other cellular component in a manner that still preserves its structures. The structure of DNA is double-stranded helices that made up from the monomer of nucleotides. Each of the nucleotides composed of three parts that are the phosphate group, deoxyribose sugar backbone and also nitrogenous bases (Adenine, Guanine, Thymine and Cytosine). This nitrogenous base are arranged in sequence and holds the information and coding for controlling the physical trait that we all have as well as the regulation of our body.
DNA extraction is simply a process that results in separation of DNA from the cells or viruses that are hosting it. Through the meaning is simple but the process is not. When we go into the peculiar details of DNA extraction, we realize that it’s more of an initial stage in other intensive DNA testing processes. DNA test could be performed for any reason; however for any DNA testing to happen the first stage normally is the isolation and extraction of DNA molecules from the cells that they reside in.

    Lichens are a sybiotic association of fungus (mycobiont) and algae (Pediastrum boryanum . The fungus engulfs the algae, then supplies carbs, nitrogen, and vitamins while the algae photosynthesizes. This unusual relationship enables the lichen to grow in environments where neither fungus nor algae would normally be able to solely survive. They are found on every continent, ranging all the way from the freezing Arctic to the sweltering grasslands of Africa. Even though they can live in such extreme conditions, they do require clean air. This is why they are excellent environmental indicator. An indicator species is an organism whose presence or absence reflects the overall health of an environment. Lichens flourish where there is good air quality; however, they cannot survive where the air is polluted and toxins loom. This is because they have no stomata, or pores, to shut to keep the toxins out; therefore, absorbing all of the pollutants in the air. Because they are such unique organisms, they accumulate different metal anions in the air than other organisms, like plants do, for example. Lichens attract toxic metal ions. An ion is a charged atom. If the ion has a positive charge, it is a cation. An ion with a negative charge is an anion. During the ion exchange process, Hydrogen, an anion, is released and metal cations move in. This process occurs when lichens are exposed to polluted air. When the lichens are dropped into any of the metal solutions, their cell wall releases the methylene blue ions, which are replaced by metal cations. Upon their detachment, the ions integrate into the solution, which in turn, colors it blue. The darker the solution, the more metal cations taken in by the lichen.     There are three specific steps required to isolate DNA from its cellular contents. The steps used to remove and expose DNA from its cell are: breaking down the food type you are using by crushing it, for example a banana or strawberries, exposing the substance to a sodium chloride (NaCl) solution, subjecting the product to detergent solution (dH2O), filtering the solution and lastly, the addition of ethanol. When beginning with a solid substance, such as a banana, crushing the substance allows for breaking down the cell membrane and cell wall, if using a plant cell. This ultimately, removes the contents of the cell from the deoxyribonucleic acid (DNA) and further exposes it for the detergent solution. After this step, one would use NaCl solution to stabilize the DNA, since we know that it is negatively charged, due to the phosphate functional group. The way this occurs is when NaCl is added to the solution the molecule dissolves and forms ions. As a result, the positively charged ions neutralize the negative charge on the DNA molecule ("Discovering DNA" 20 Feb. 2014). The technique used to separate the DNA is by adding a detergent liquid to the solution, which allows for the cell membrane to break down. This step specifically emulsifies the phospholipids and proteins that make up the cell membrane ("Discovering DNA" 20 Feb. 2014), in order to further expose the DNA. Then in order to separate the DNA from the proteins and lipids one must use a filter paper with an appropriate pore size so that the flow rate of the solution is effective. Once the flow rate is minimal, the excess in the filter, consisting of cell debris and banana residue, can be disposed and then the filtrate is focused upon. With the specific amount of…
