Microbes in the Environment

Abstract
Microorganisms are plentiful and widespread in the environment. In this lab, we undertook to determine the differences in the agars being used and the different colony count observed. After taking four different samples of microbes from the environment and swabbed them in two different plates one with nutrient agar and the other with sabouaud dextrose agar. After the microbes had incubated for 48 hours no results were discovered from the swabs we had taken from the environment. This lab further investigates the different swabs from the environment other group got and the different results discovered from the agars being used and the colony count observed.
Introduction
The natural environment is populated with many microorganisms suspended in the air, in water, and associated with other organisms. All surfaces in the lab and in the environment are potential source of contamination. In order to examine microorganisms without the use of a microscope, they must be given the proper condition and nutrients to flourish into colonies. In this lab, microorganisms were taken and cultivated from four different sources. For each of these, there were two different medias used – nutrient agar and sabouraud dextrose agar. The intent is to determine the differences in the agars being used and the different colony count observed.
Materials and Methods
Initially, two plates were gathered one was nutrient agar (NA) and the other was sabouraud dextrose agar (SDA). Then gathered two packs of cotton tipped applicators. Then a magic marker was used to label each plate making quadrants numbering each one. Also the name and date was written on the edge of the plate. In the NA plate Whitney swabbed her left index finger in the first quadrant. Then washed her hands. In the second quadrant Whitney took her clean left index finger and swabbed it in there.     Paul Fitts’ was the first to discover the relationship between the speed of movement and accuracy requirements. This has become one of the most fundamental principles of motor control. Fitts claims a relationship between task difficulty and movement time. He quantified task difficulty as “index of difficulty” which consists of the ratio of twice the amplitude over width of the target (2Amplitude/Width).     The tooth formula is a shorthand technique that biologist use to keep track of the teeth on a particular jaw. The formula consisted of two lines, top jaw and bottom jaw, which were used to count how many incisors, canines, premolars, and molar teeth a species had. Species A, Odocoileus virginiaus (white-tailed deer), had a tooth formula of 0033/3133. On the top jaw, the deer had zero incisor and canine teeth but have three of both the premolars and molars. On the bottom jaw, the deer had three incisors, one canine, three premolars, and three molars. Species B, Canis latrans (coyote), had a tooth formula of 3142/3143. On the top jaw, the coyote had three incisors, one canine, four premolars, and two molars. On the bottom jaw, the coyote has three incisors, one canine, four premolars, and three molars. Both the deer and coyote skull were long and skinny towards the nose and mouth. The coyote skull had a much rounder nose than the deer.     There is also a lot of variation among microorganisms. I found many different shapes, sizes, colours and indeed colonies of microorganisms. This means that microorganisms are abundant in our everyday lives. They exist on absolutely everything; I found them on the floor and inside my cheek. As I previously thought, upon macroscopic examination, there was more bacterial growth on the floor than inside my mouth, although I did expect to find some more variation in my mouth as there were only colonies of the same kind present. There was much more variation in the types of microorganisms present on the floor samples. Perhaps this could have been due to the fact that some bacteria do not grow on artificial media. I feel that if I had of used different types of agar plates that were the favoured media for bacterial growth in the mouth, I could have found more bacteria present. Another reason for the small amount of growth could have been due to experimental error when obtaining the samples with the swabs. I could have swabbed with one side on the swab and smeared the other side of the swab on the agar plate which would not have come into contact with the area to be examined such as the floor. From my results I gather that some microorganisms favour certain agar plates, for example I only found yeast growth on the SDA plates as this…
