Target Factor Analysis Lab Report
Objective: The goal of the experiment was to use UV/VIS spectroscopy to collect data to quantify acetaminophen, aspirin, and caffeine from a migraine tablet where there is heavy spectral interference using Target Factor Analysis1. The migraine tablet has other ingredients in it like FD&C blue #1, stearic acid, and saccharin. Saccharin is the ingredient thought to have the biggest interference and was used as the experimental basis2.

Instruments:
UV/VIS spectroscopy was used to analyze the absorption spectra of the many solutions in this experiment. It was chosen for its ability to collect absorption data quickly and efficiently. A Target Factor Analysis program was used to treat the UV/VIS data collected. Target Factor Analysis was used as the calculation method instead of CLSR due to it taking into account the interference2.

Discussion: Table 1 is the calibration matrix

This is one of the reasons why TFA was used and why there is some error in the calculation of the concentrations. Error also could have been caused from reference cuvette of 0.01 M HCl used in the spectroscopy absorbing the light energy over multiple runs. Figure 2 shows all calibration spectra (color of your choice) and the four unknown spectra. Table 4 shows max and corresponding absorption coefficient of pure caffeine, aspirin, and acetaminophen. Caffeine has the largest absorptivity coefficient of 2637.202158 and Acetaminophen and aspirin have considerably lower coefficients of 1086.751132 and 1257.296755 respectively. This data has a correlation with the solubility, structure, and molar mass of the compounds. Caffeine has the largest molar mass and the solubility of caffeine at 25 oC is 2g/100 mL, the highest of the three analytes. This difference in solubility is due to the polarity of the structure of caffeine when compared to the polarity of the structure of the other

Our Infrared Spectroscopy (IR) spectrum for our Acetaminophen KBr pellet provided a handful of strong peaks and hints that helped us to hypothesize what functional groups might be in Acetaminophen. The peaks that were at the highest wavenumbers were analyzed first. The strong peak at 3325.82 cm-1 was an indicator that there was stretching occurring between a Hydrogen and Nitrogen atom. This signified that Acetaminophen could contain an amide group. The next defined peak is located at 3162.26 cm-1, and the section between these two peaks has a slight broad descend.…

Hence, from least to greatest relative polarity, the order of the compounds are as such: ibuprofen, aspirin, acetaminophen, caffeine. According to their molecular structures, caffeine is the most polar because of the unpaired electrons on the nitrogens and the unpaired electrons on the oxygens. In other words, one side of the molecule has two amides and is more negatively charged due to the lone pairs of electrons on the two nitrogens and the two oxygen (6 pairs of lone electrons), while the other side has 1 amine and is less negatively charged. On the other hand, aspirin is only slightly polar in comparison to the other compounds. This is because it has an ester group and carboxylic acid group; however, only the oxygen in the carboxylic acid has lone pairs of electrons that can form bonds.…

It is important to note that acetic acid is the non-polar component of the experiment. On the other hand, the stationary phase corresponded to the Silicon on the TLC plate. From the dipole in Silicon and Oxygen bonds, this is regarded as the polar component of the experiment. Compared to the mobile phase, the stationary phase results in the polar functional groups to be a stronger attractant. As a result, the most polar functional groups will travel up less the TLC plate. Due to the non-polar compounds not sticking to the stationary phase as strongly as the polar compounds do, non-polar compounds should have a larger Rf value than polar compounds. As previously stated, the least to most polar is Ibuprofen, Acetylsalicylic Acid, Acetaminophen, and Caffeine respectively. This shows that ideally, Caffeine has the lowest Rf value while Ibuprofen should have the largest Rf value. In the Observations section, we calculated that Ibuprofen’s Rf value is .81, Acetylsalicylic Acid’s Rf value is .64, Acetaminophen’s Rf value is .46, and Caffeine’s Rf value is .10. This illustrates that the Rf values trend match the the four analgesics’ polarity…

We found the Rf value by the distance spot travels divided by the distance the solvent travels. From the TLC samples collected, the solvent travels to be 7.7 cm and the distance the spot travels from aspirin, acetaminophen, ascorbic acid, and unknown to be 7.0 cm, 7.7 cm, 7.6 cm, and 7.5 cm respectively. Since the two…

The goal of this experiment was to use TLC Analysis to determine the analgesic drug present in the unknown solution 154. One observation of the unknown spotting solution was that it was cloudy. After forming the unknown spotting solution it was then heated to make it a more saturated solution thus enabling the spots on the TLC plate to contain more of the drug making it easier to compare it with the other drugs. After the TLC plates were developed and visualized first through UV light and then through an Iodine chamber ( though no more spots were visualized with iodine chamber) , Rf values were then taken of each spot on the plate. The Acetaminophen had a value of 0.323, Aspirin had a value of 0.597, Caffeine had a value of 0.081, Unknown 154 on the first plate had 3 different values; 0.081, 0.306, 0.597, Ibuprofen had a value of 0.698, Salicylamide had a value of 0.587, and Unknown 154 on the second plate had three different values as well; 0.079, 0.397, and 0.587.…

The main purpose of this experiment was to utilize the principles of cyclic voltammetry to determine technique accuracy in calculating the actual concentration of an acetaminophen elixir. Five standard solutions of acetaminophen and an acid buffer solution were created and analyzed with a potentiostat to determine the peak anodic voltage and current during the scanning process. Three solutions of Tylenol®—containing acetaminophen—and acid buffer were then created and scanned at the same scan rate as the five standard solutions. Additionally, the most concentrated standard solution was scanned at five different scan rates (between 100 and 500 mV/s) in order to develop a relationship between observed current and scan rate. The Randles-Sevcik…

The UV/Vis spectrophotometer work by passing the light by strong lamp through a monochromator, which split the light into its component colours using a gratings. Then, the desired wavelength of light beam is direct through the sample compartment which the cuvette is placed by using the mirror and slit. At other side of the compartment, there is a detector which can determine the amount of light beam pass through the solution in the cuvette, thus the absorbance of the solution in the cuvette can be determined. In addition, the absorbance of light is determined by three factors which is the nature of the solution, concentration of the solution, and the distance of light travel through the solution (path length) which stated in the Beer-Lambert Law.…

This experiment aims to determine the acid dissociation constant value of Methyl Red by means of spectrophotometry. Ten samples were spectrophotometrically analyzed under a UV-Vis spectrophotometer and from the absorbance values acquired, calibration curves were constructed to determine the concentration of the species of interest particularly HMR(acidic methyl red) and MR-(basic methyl red). The principle of Beer’s Law was used since it establishes the direct proportionality between the absorbance and concentration.A pH vs. log(MR-/HMR) curve was also constructed to discern the pKa which is the y-intercept of the equation of the line. A pKa value of 4.9414 was obtained .By comparing the experimental value to the literature value of pKa which is 5.00, a 1.172% deviation was calculated.Based on the aforementioned results, it was confirmed that spectrophotometry is a feasible technique in the determination of the acid dissociation constant of a methyl red .…

the results calculated for green and black tea were much closer to the accepted value than all the comparable results calculated for green and black tea . Black tea had the most caffeine, and the green tea had the least. This is make sense, as usually the black tea has more caffeine in it, as black teas are fermented making them have more caffeine in them. This can be seen from table 2 in which the black tea had a higher melting point range of 204.7-241.7 ˚c, while black tea had 191.3-239.9 ˚c. However, there is source of error that may have contributed to the 2. 85% and 2.05 %. For this experiment could have been from mishandling the equipment, not weighing the correct amount of tealeaves, allowing other substances in, or not performing the extraction…

The fact that each compound has its specific emission maxima without overlap from the other, allows their quantification without prior separation when they are found in equal ratio. However, when one drug is minor this method failed to determine the minor component. Therefore, the first derivative technique was chosen for simultaneous determination of both VAL and SAC in synthetic mixtures of different ratios whether they are equal or one is minor and the other is major. Different excitation wavelengths were examined to reach the most suitable one. Using the selected λex, both VAL and SAC gave a reasonable relative FI at λem 416 and 314 nm, for Val and SAC respectively. Another λex for VAL was reported [17] at 227 nm. Although this wavelength λex 227 gives higher response for VAL than λex 249 nm, it couldn’t be used as it yielded a very low and non-reproducible RFI values for SAC which will hinder the simultaneous determination of both drugs in their dosage form…
