Uv-Vis Procedure
PROCEDURE
1) A standard was prepared in solvent and was placed into sample holder for the purpose of calibration curve.
2) The liquid sample was also prepared in a similar manner and placed into sample holder.
3) The UVProbe Software was opened and the method(M) was set to wavelength of 800-0.0 at medium speed for measuring liquid sample absorbance , and the method was saved in a file according to sample name.
4) The standard for baseline measurement was run. (The first run using an empty cuvette followed by the standard sample)
5) Sample measurement was continued.
6) All the results were saved and the spectrum produced was saved.
7) The main component in UV-VIS unit along with the diagram of the unit was drawn.
8) Spectrum obtained was analyzed based on wavelength peak and compared against calibration curve.
9) The analyte concentration found in the unknown was determined.
10) All the findings were summarized based on the spectrum obtained and related information derived from the experimental work and from references.
STANDARD OPERATING PROCEDURE FOR UV-VIS UNIT
Start-Up
A. Equipment
1. The power supply was connected to the unit.
2. The unit and PC was switched on.

B. Steps to Perform Analysis
1. The UVPorbe Software was opened and CONNECT button was clicked. A series of tests is automatically conducted which may take about 5 minutes. All checklist is made sure to be green.
2. Prior to analysis , the method was set according to requirement, by clicking the M button.
3. The method was then saved based on sample name and the FILE menu was clicked. The method is then saved as on a created new folder.
4. Calibration must be done using BaSO4 for solid sample or solvent for liquid (50%ethanol and 50% DI water / 70% ethanol and 30% in DI water) containing the analyte (five concentration of analyte plus bank i.e without sample) as standard calibration.
5. The BaSO4 for solid or solvent for liquid cell was placed at sample holder. The BASELINE icon

The samples were placed in a desiccator instead of being left to cool on the lab bench between each heating. This is because the beaker and contents would cause the mass of fluctuate when placed on the tared balance if not cooled in the desiccator. This is used to store the beakers and the anhydrate so that the anhydrate does not absorb moisture from the air.…

Initially the plates were rinsed with ethanol, then dried with lint0free tissues. Next 1-2 g of benzoic acid were transferred to a mortar and a pestle was then use to grind the sample. About 100-150 mg of oven dried potassium bromide were added into the mortar and mixed well and then grinded. Next a screw was attached to a bolt and tightened together to make a sample holder. Next another screw was attached to the bolt to fasten it. The sample holder was then mounted onto the sample holder support on the table bench and a wrench was used to tighten the screw. It was held there for about 1-2 minutes and the bolts were loosened and removed. It was made sure not to fasten the bolts too tight or it would damage the bolt and the screw sample holder. After removing the two screws and transparent solid sample pellet ready to be measured was found formed within the screw.…

The purpose of this experiment was to determine the concentration of an unknown amount of copper sulfate using the spectronic 20. Four solutions of different concentrations of H₂O and CuSO₄ were put into the spectronic 20 to observe their absorption of light. Then, a graph was created to plot down the measurements. Using the calibration curve of the graph, the concentration of the unknown substance was found out to be .12M.…

Calibration is the process by which a stated measure such as the volume of a container is checked for accuracy. In general, measurements of mass can be determined more precisely and accurately than measurements of volume. Therefore, the mass of the liquid contained or dispensed by the glassware will be measured and the corresponding volume calculated using the density of the liquid. However, a relatively small change in temperature causes a change in the liquid’s volume and thus its density.…

Analytical chemistry involves the separation, identification and determination of amounts of components in a particular sample. Accuracy and precision are important in data acquisition to be able to acquire substantial results in the end of the experiment. To attain such goal, replication of measurements must be performed. The reliability of the measurements may then be evaluated through various statistical parameters [1].…
