    An unknown bacterium was handed out by Dr. Honer. The appropriate tests were prepared and applied. The first procedure that was done was the gram stain. Under a microscope, if the gram stain is purple, the bacterium is gram positive, if the stain is red, it is gram negative. The next test was the fermentation tests for glucose, sucrose and lactose. If an organism is able to ferment the carbohydrate, there will be acid produced, which will turn the broth yellow. Homofermentation shows that only one product was produced-acid. Other organisms will carry out heterofermentation, the presence of acid and gas. If the organism is unable to ferment the sugar in the tube, there will be no change in the orange/red color of the broth and will be considered alkaline. The following tests that I performed were tests for methyl red and vogues proskauer in SIM media. A positive methyl red test indicates a mixed acid fermentation and would turn the organism’s broth red, if no color presents, it is negative. A positive vogues proskauer test indicates fermentation to neutral products and will turn the broth red, if no color presents, it is also negative. Stab inoculating the SIM media for these tests will also show if the organism produces sulfur or indole and if it is motile. If sulfur is present, then the medium will be black, if it produces indole, the medium will be red, and if it is motile, there will be growth throughout the tube. I also grew the organism on MacConkey agar. After inoculation, visible colonies may or may not be present.…

    The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped. After determining the unknown was gram-negative, other bacteria’s were able to be eliminated from the options. The Triple Sugar Iron Test was chosen next because the unknown could be tested for glucose fermentation, lactose fermentation, sucrose fermentation and sulfur reduction. If the organism is able to ferment glucose and lactose and/or sucrose it will turn the medium yellow throughout (Leboffe & Burton 329). When the Triple Sugar Iron Test was complete, it showed a yellow butt, yellow slant and gas production. This determined that the unknown ferments glucose and one or both of the other sugars. The gas was produced by fermentation. The color change in the agar brought the unknown down to two options of unknowns, Escherichia coli or Hafnia alvei. The SIM medium test was chosen next to see sulfur reduction, indole production from tryptophan…

    For unit 7.3 the experiment needed 2 microfuge tubes one labeled "L" for lactose and the other labeled "M" for maltose. The experiment also called for a total of 3 clean plastic pipettes. With one pipette add milk up to the 0.5 line of the lactose tube. With the second clean pipette add maltose solution up to the 0.5 line of the maltose tube. With the third clean pipette, add lactase solution to each tube, until the level of mixture in each tube comes up to the 1.0 line of the tube. Place both tubes in the 40 degrees celsius water bath and incubate them for 10 minutes. The other thing needed is glucose strips. After the tubes have been incubated for 10 minutes, place a glucose strip in each tube for one second. Let it sit for thirty seconds, then compare to the chart provided. For unit 7.4 the experiment called for two microfuge tubes. One labeled "control" and label another one "EDTA". Add 1.25 mL of EDTA to the EDTA tube, add 1.25mL of distilled water to the control tube. Add 3 drops of milk to each tube, invert the tube and let sit for 1 minute. Then add 3 drops of lactase solution to each tube. Then place both tubes in the 40 degrees celsius water bath and leave them for 10 minutes.     When I received my slant culture, I immediately made a streak plate and started the Gram Stain protocol1. In the first two slides, I made and stained, my results were ambiguous. Without putting the slide into the microscope, I noticed one was pink and the other purple. To resolve it, I discarded my slides and started again and was extremely careful in proceeding the Gram Stain. I concluded that my unknown A was gram-negative bacilli. Using the dichotomous key for gram-negative bacilli I started the protocol for the lactose test2 which would help me determine which test to conduct next to narrow down list of possible bacteria. I conducted the protocol for MR test3 and citrate test4 the same day as the lactose test to speed up the process of elimination. With conducting the MR.
