Xylanase Reaction Lab Report
The effect of temperature on xylanase activity was determined by incubating the enzyme in 0.25 ml 1% (w/v) birchwood xylan at temperatures between 40 and 70 °C with regular interval of 5 °C. Thermo stability was determined by incubating the enzyme at temperatures between 55 and 95 °C with regular interval of 10 °C. Moreover, thermo stability was also investigated at room (24 °C) and ice temperature (4 °C). Xylanase residual activity was calculated after different times. After incubation the reaction was stopped by adding DNS reagent and absorbance was measured at 540 nm.
The effect of pH was determined by measuring its activity at various pH value using different buffers such as sodium citrate (pH 4-6) and Tris-HCL (pH 7-9) each at 0,01 M.
