Understanding how catalase activity is affected by pH is the purpose of this experiment.
Introduction
Enzymes play an important role in daily life because of the chemical reactions. Almost chemical reactions require the presence of enzymes to promote the metabolic process. They are known as the incredibly efficient and highly specific biological catalysts. Most enzymes are protein with the ability to enhance the rate of reaction between molecules. To catalyze a reaction, the enzymes have to create the best environment that is called active site. The active site has to be fitted with the reactant or substrate for the enzyme’s reaction. Each tube was filled to the 1 cm mark with catalase. After that, to the 3 cm mark, while test tube 1 was filled with HCl, test tube 2 was filled with distilled water, and test tube 3 was filled with NaOH. Then, pH was immediately measured on each tube and recorded. After 5 minutes, each tube was added hydrogen peroxide to the 7 cm mark and swirled to mix. After 20 seconds, the height of the bubble column of all tubes was measured in millimeters and recorded.
Results
Tube 2 with the pH of 7 had the highest height of bubble column at 10 mm. However, the tube 1 with pH of 1 and tube 3 with pH of 10 did not create any bubble so their bubble heights were …show more content…
That means catalase was affected by the change of pH. Catalase could only work well with the neutral and stable pH value. The high or low pH values generally led to the loss of activity of this enzyme. The optimum pH of enzyme is known at the point where the activity of enzyme is the best. It is also considered the most favorable pH value of enzyme. Therefore, the optimum pH of catalase was pH of 7.
The term “denatured” is used for the enzyme has been changed its nature or natural quality. Therefore, the enzymes resulted in the completely loss of activity are recorded as “denatured” proteins.
The next experiment would test if the temperature can promote the denatured enzymes to catalyze again. All of the above tubes would be placed in boiling water and recorded the height of bubble column after 5 minutes. The hypothesis tested is that the enzyme cannot recover to its natural quality.
