LABORATORY REPORT
Activity: Enzyme Activity
Name: Angela Collins
Instructor: Catherine Rice
Date: 07.09.2014
Predictions
Sucrase will have the greatest activity at pH 5
Sucrase will have the greatest activity at
70 °C (158 °F)
Sucrase activity increases with increasing sucrose concentration
Materials and Methods
Effect of pH on Enzyme Activity
Dependent Variable amount of product (glucose and fructose) produced
Independent Variable pH Controlled Variables temperature, amount of substrate (sucrose) present, sucrase + sucrose incubation time
Effect of Temperature on Enzyme Activity
Dependent Variable amount of product (glucose and fructose) produced
Independent Variable temperature Controlled Variables pH, amount of substrate (sucrose) present, sucrase + sucrose incubation time
Effect of Substrate Concentration on Enzyme Activity
Dependent Variable amount of product (glucose and fructose) produced
Independent Variable amount of substrate (sucrose) present
Controlled Variables temperature, pH, sucrase + sucrose incubation time
4. Describe what is measured as an indicator of sucrase activity and why this is an indicator of sucrase activity. the optimal density is measured because it measures the amount of glucose and fructose produced.
5. Explain why denatured sucrase was used as a control. because denature sucrose will not have as much activity as the other sources.
Laboratory Report/ Angela Collins/ Enzyme Activity/ Catherine Rice/ 07.09.2014/ Page [1] of [4]
6. Addition of DNS at the end of the incubation period stopped the reaction by denaturing sucrase. Explain why it is important to denature sucrase before measuring product concentration.

o If all four bacteria were able to metabolize glucose, explain why all four were not able to metabolize lactose and sucrose. Consider the different chemical structure of each carbohydrate and what the bacteria would need to metabolize each carbohydrate. The bacteria quickly metabolized the glucose, initially producing an acid slant and an acid butt in a few hours. After further incubation glucose is consumed because the bacteria could not use lactose or sucrose.…

    1) The purpose of this lab was to determine the rate of enzyme activity under variety of different conditions, such as, different amount of drops of enzymes and different temperature of water. The class measured the pressure in the test tube during the reaction of the substance with, 1.5 ml of H2O2, 1.5ml of H2O and different amounts of enzyme drops, to determine how much oxygen gas is produced during the reaction since the pressure of the test tube will get higher as more oxygen gas is accumulated during the reaction.…
