Gram Stain Lab Report
When I received my slant culture, I immediately made a streak plate and started the Gram Stain protocol1. In the first two slides, I made and stained, my results were ambiguous. Without putting the slide into the microscope, I noticed one was pink and the other purple. To resolve it, I discarded my slides and started again and was extremely careful in proceeding the Gram Stain. I concluded that my unknown A was gram-negative bacilli. Using the dichotomous key for gram-negative bacilli I started the protocol for the lactose test2 which would help me determine which test to conduct next to narrow down list of possible bacteria. I conducted the protocol for MR test3 and citrate test4 the same day as the lactose test to speed up the process of elimination. With conducting the MR. Citrate and Lactose test all in the same day, I learned that time management was a big deal and I learned quick and caught up with the class. The next class I observed my lactose tube and my results were AG, yellow with bubbles. The …show more content…
K.phumoniae is not airborne but this bacterium is easily spread in hospitals and it can be caught by anyone who comes into contact with the infected person’s feces if proper precautions are not taken. It is recommended that if anyone needed to enter a room with a patient that has a K.phumoniae related illness, to wear a gown and gloves. As for the patient, it is recommended to wash their hands profusely to prevent any chance of anyone else contracting the illness. Some, not all K.Phumoniae are highly resistant to antibiotics. K.Phumoniae that produces an enzyme called carbapenems it doesn’t allow the antibiotics to destroy the bacteria and treat the patient and to be able to determine which antibiotics will work has to be determined by the doctor. The K.Phumoniae that is found in the gut can be treated with

Since P. aeruginosa is an opportunistic organism, most of the people that are affected by it are people whose immune systems are compromised, which is why P. aeruginosa is such a big problem in hospitals and clinics. The best way to control this pathogenic organism is by proper isolation procedures, aseptic technique, and careful cleaning of equipment. Using aseptic technique and cleaning all surgical equipment properly is very important because P. aeruginosa is easily spread in hospitals by nurses, surgeons and other hospital…

Observations: The color changing process took longer than we had expected. We observed that after putting the penny in the beaker, the zinc powder surround the penny, which helped it changed its color. We couldn’t complete the experiment so when we took the penny from the beaker, the change in the color wasn’t completed. There were some dark spots and some light parts.…

Prokaryotes are a large group of organisms with no membrane bound organelles. They consist of two domains: Archaea and Bacteria. These organisms are only found in extreme environments such as volcanoes. Prokaryotes are still being researched and are a very diverse group. In this lab we focused on trying to identify if the bacteria found had a lot of peptidoglycan by gram staining. Testing this could be done by using a Petri dish full of agar and testing different bacteria on it to see if the bacteria obtained is gram positive or gram negative. My hypothesis is there will be a lot of bacterial growth on all of the plate.…

In this lab, we will take a trip to the planetarium lab and will learn about Right ascension and declination, and altitude and zenith. After looking at the different points shown, we will log the altitude and zenith in the chart in our lab manual. Now we will look at the same points and label the right ascension and declination. Then we will learn about the easiest way to locate the star Polaris.…

The cold pressor test is done by having the subject submerge their hands in a bucket of cold water to determine cardiovascular changes such as blood pressure and heart rate. For this experiment we wanted to know how long participant can keep their feet in the bucket of ice water before they experience any pain while looking at neutral and positive pictures and also rate the intensity of their pain. For this reason we hypothesized that, if participant submerged their feet in a bucket of water while looking at the positive stimulus, it would take them a long time to experience pain and will keep their feet in the bucket of ice for a long time than when participant are looking the neutral stimuli while they have their feet in the bucket of ice, we also hypothesized that heart rate will increase the longer participant keep their feet in the ice water. The null hypotheses was that, if participant submerge their feet in a bucket of water while looking at positive stimulus, it would take them the same amount of time to experience pain as when they are looking at the neutral stimulus and that heart rate will stay the same throughout the experiment…

The lactose test appeared pink still, which indicated no fermentation had happen and no gas was found either. The sucrose test appeared yellow indicating that sucrose was fermented turning the tube acidic and surprisingly no gas was found. After these tests and with the red growth on the TSA media it was confirmed that it was indeed the bacteria Serratia marcescens. In the gram-positive bacteria tests the MSA media turned completely yellow, which indicated mannitol was fermented turning the plate acidic. The glucose-bromocresol purple tube appeared completely yellow as well indicating glucose was fermented which turned the agar acidic. The bacteria Staphylococcus aureus was identified because it's the only gram-positive bacteria, which is able to turn both MSA and glucose-bromocrosel tests acidic, which gives them their yellow appearance. No further testing was needed to identify the two…

In unit 7.3 the experiment tested the ability of lactase to specifically bind and interact with lactose compared to maltose. In unit 7.4 the experiment tested the role, if any, that metal ions have on the activity of lactase. My hypothesis for unit 7.3 was knowing that lactase is specific for lactose, lactose will separate into galactose and glucose, as maltose will not change (153-155). Lactase should like lactose. For unit 7.4 my hypothesis was that EDTA will remove the ions, and will not work.…
