Question: Is there a subsequent effect of the bacteria when decolorizing with alcohol?
Hypothesis: If there is either a removal of the outer cell membrane or dehydration of the bacteria then there is to be an effect in cause of the decolorizing agent .
Materials: One bunsen burner, Two bacteria cultures, Two microscope slides, Inoculating loop, Clothespin, Gram’s crystal violet, Gram’s iodine, Decolorizing agent: Gram’s alcohol, Safranin, Distilled water, 1 large petri dish, 1 small petri dish, Microscope
Procedure: Once in groups and materials are obtained one is to heat fix a bacteria culture by sterilizing the inoculating loop with an ignited bunsen burner using the loop to obtain the distilled water to place on a microscope slide the inoculating loop is then again sterilized and used to place a single bacteria culture unto the same microscope slide. The microscope slide is then set a side attached to the clothespin to semi air dry where once done the slide is then passed through the ignited bunsen burner several times to heat-fix the observed smear produced. The bacteria is then …show more content…
When conducting the gram staining test there was of evidence to support the initial hypothesis of the effect of the bacterial cell with the addition of Gram’s alcohol as the the lipopolysaccharide layer was removed in Culture A and in Culture B there was in observance of a shrinkage in pore
