1. Introduction
Lonicerae Japonicae Flos (LJF), an important traditional Chinese medicine herb, has been widely used for centuries in many prescriptions to treat acute fever, headache, respiratory infection, and epidemic disease via its anti-inflammatory (Cheng et al., 2014), antibacterial (Shan et al., 2007), and antioxidative activities (Fang et al., 2015). Many studies shown that LJF also offers anti-diabetic (Zhou et al., 2016), and immunomodulatory properties (Kao et al., 2015). Chlorogenic acid (CA, Fig. 1) is one of the major ingredients in LJF and has been used as a marker for the quality control of LJF in Chinese pharmacopoeia. CA-SMEDDS formulation (1 g) was dispersed in distilled water (100 mL) and mixed by slightly stirring at 37 °C until microemulsion (0.6 mg/mL of CA) was obtained. Proper amount of microemulsion was filtered and 0.2 μL of them was deposited on a film-coated copper specimen grid and allowed to stand for 2 min, then any excess fluid was removed by clear filter paper. CA was eluted against a C18 column (Plastisill, 5μm, 4.6 mm×250mm; Dikma technology co., LTD, China) and guarded with a precolumn at 25 °C. Mobile phase consisted of 15% acetonitrile and 85% phosphoric acid solution (0.1%, v/v). The injection volume was 20 μL and signals were collected at 327 nm at a flow rate of 1.0 mL/min. Method validation assays were carried out by selectivity, linearity, precision, recovery, and stability, respectively. CA was separated well from endogenous materials in dissolution media, blood, and each tissue. Separate standard curves were used for each experiment (R2>0.9994). The inter-day and intra-day coefficients of variation in all experiments were less than 9%. The recovery of CA was in the range of 80.67%-93.75%, and good stability was obtained within 24 h (RSD
