Peroxidase Enzyme Lab Report
In this lab the peroxidase enzyme is tested in a dormant avocado seed as well as an avocado seed undergoing the process of germination. A gas pressure will be used to test the seeds and see if the peroxidase enzyme is present in either of the seeds. A catalyst is very similar to track spikes. Spikes increase a runner’s speed, as a catalyst speeds up the chemical reaction time in a plant. Neither the catalyst nor shoes are changed in these actions. Enzymes are macromolecules that act like catalysts. A catalyst will lower the activation energy within a seed, which will allow the reaction to go quicker. An avocado seed is used in this experiment. For this type of seed to start the process of germination, the favorable climate is typically very warm. If someone was to plant an avocado seed and see the quickest results, they should place the seed in the sun. Along with waiting a few days between watering the seed, allowing the soil to dry out. For the seed to germinate properly, the bottom half of the seed will be emerged into water. Once the top of the seed becomes dry it will crack and the exterior brown skin will come …show more content…
A dormant avocado seed and an avocado seed in the process of germinating will each be tested, using a gas pressure. This tool will record if the peroxidase enzyme is present in either of the seeds. The seed undergoing germination will have awakened enzymes however the dormant seed will have un-hydrated enzymes. Depending on the amount of gas released from each seed will determine how much catalyst activity is going on in the seed. The independent variable is the amount of time that the seed will be tested for gas. The dependent variable is the gas pressure and how much gas is released from each seed. The peroxidase enzyme catalyzes the change form hydrogen peroxide into water and oxygen gas. It’s safe to assume that the dormant seed will not produce as much gas as the awakened seed

The main objective of this lab was to observe the activity of enzyme peroxidase in real time under different experimental conditions. To see how peroxidase reacts with its substrate guaiacol under different conditions. We measure the amount of substances per test tube and then combine all of them together. Each test tube contains a measured amount of any listed substances including H₂O. Each measured amount of peroxidase along with its substrate guaiacol, and other listed substances will show how actively the enzyme oxidizes the substrate. To measure the…

The purpose of this experiment was to inspect the effect of environmental conditions on hydrogen peroxidase. The three conditions tested were the effect of peroxidase concentration on the rate of the experiment, the effect of pH of the rate of peroxidase activity, and the effect of temperature on the rate of peroxidase activity. During the lab, the lab group tested 7 test tubes, including 1 blank, with different amounts of pH 5 buffer, H2O2, Peroxidase, and Guaiacol. After the certain amount of mL per substance was mixed, the absorbance readings for the effect of peroxidase concentration were taken from the spectrophotometer. The results for the effect…

Next, 50ml of distilled water will be measured using a graduated cylinder. The 50g of catalase and the 50ml of distilled water will be put into a blender and blended until liquefied. The catalase solution will then be introduced to a Buchner funnel filtration system to separate the catalase extract from any solid particles left over from the blending process. This step will be repeated until all of the catalases have been blended and filtered. Next, 5 ml of hydrogen peroxide will be measured out using a graduated cylinder. It will then be poured into a small Erlenmeyer flask. This will be repeated five times, representing the five trials for each catalase treatment. Once the Erlenmeyer flask has been given 5ml of hydrogen peroxide, 10ml of the S. oleracea catalase will be measured out and poured into the Hydrogen peroxide filled flask. When the catalase and Hydrogen peroxide meet a reaction will occur and the hydrogen peroxide will be broken down into water and oxygen. The oxygen released from the reaction will be recorded by the Gas Pressure sensor. This step will be repeated five times for each necessary trial, then the next catalase extract will be…

In our experiment we conducted an experiment to validify our hypothesis: “ If the enzyme concentration increases, it would alter the rate of which the color changes.” We hypothesized that the increase of an enzyme concentration would result in the increase of reaction rate; our experiment provided enough data to prove our hypothesis. The function of this experiment was to investigate how the influence of turnip peroxidase enzyme on the rate of reaction. As a given we would have 20g of turnip in 500 mL of deionized water. We would have a three different trials and in those trials we would do it three times.…

The results of our experiment showed the solutions in both tube 1 and tube 2 increasing in absorbency in the first eight minutes but then tube 1 continued to increase while tube 2 began to balance out. Tube 3, our blank, managed to stay at 0nm the entire twenty minutes. From this data, we can conclude that our hypothesis was supported that EDTA had a greater change in absorption over PTU.…

The enzyme dramatically speeds up reactions by lowering the activation energy barrier. Every enzyme has a unique shape because the substrates that bond to it are all different shapes. It fits like a lock and key. Once the substrate enters the active site, the enzyme slightly changes shape in order to lock in the substrate and holds it in place. Enzymes allow organisms to live by increasing the rate of a chemical reaction. The peroxidase enzyme catalyzes the reaction of the formation of oxygen. Various temperatures and pH levels will be tested in order to determine the optimal environment for the peroxidase…

Four environmental factors of enzymes were tested in lab. The changing of pH, substrate concentrations, temperature, and an inhibitor (NaCl) and the effects it hade on the enzyme turnip peroxidase. Enzymes are biological catalysts which increase reaction rates by lowering the activation energies of substrates. A substrate is a reactant that interacts with the enzyme. The enzyme and substrate can be viewed as the recently discovered "induced fit model", which suggests enzymes are flexible and dynamic things that change their shape so all these substrates (reactants) can become catalyzed when the activation energy is lowered and the reactions happen a lot faster. Sometimes in cells though it may not need a particular substrate (reactant) so an inhibitor comes into play. Inhibitors are basically regulators that inhibit (disallow) the process of catalysis to take place within a particular substrate. There are two types of inhibitors that occur in such reactions. Competitive inhibiters are remarkably similar to the substrate, so much that it can match interchangeably with the substrate, thus leading to a halt in the production of the intended product. Another variety of inhibitor is the allosteric inhibitors. Allosteric inhibitors change the shape of the enzyme by binding to a different site other the active site, which is the usual site for catalysis. Usually the allosteric inhibitors make contact with the side of the enzyme opposite the active site. In some occasions however an allosteric activator is introduced and functions by connecting to the enzyme in a way that it allows for easy access for a substrate to the active site. Allosteric activators are the opposite of allosteric inhibitors. These regulators (both inhibitive and active in function) help keep the cell in homeostasis by not allowing too much or too little of a needed or not needed product to be produced.. Enzyme activity is also regulated by cofactors which are either metal ions (e.g. Zn2+, Mg2+) or…

The enzyme peroxidase has been shown to break down H2O2. Enzymes are known to increase the rate at which a chemical reaction occurs. We looked at factors that affected the breakdown of hydrogen peroxide. These effects are the different temperatures and pH levels the enzymes were placed in. We found that the optimum, or best condition, temperature for the enzymes tested was about 22 degrees Celsius. The optimum pH level for the enzyme was 7.…

Over a two week period of time in the laboratory, we experimented and tested the reaction rate of a peroxidase enzyme and the factors that affected it, both positively and negatively. The purpose of these experiments was to probe and manipulate the activity of the enzyme peroxidase by varying temperature, pH, the amount of enzyme compared to the substrate and the effect of hydroxylamine. Peroxidase activity is expressed when the potato extract is subjected to stresses such as low temperature (El-hilali et al., 2012). The most eye catching factors that we tested for their impact on enzyme activity involved change in pH, temperature, boiling extract, and the effects of probing the active site with hydroxylamine. In the first part of…

Seeds must protect themselves until they are in the right conditions for them to grow. Until the conditions are right, seeds are dormant and appear dead and it is difficult to tell if they are actually alive (Germination). The embryo within a seed will eventually die while it is dormant if it doesn’t begin germination by a certain time. Depending on the seed, the length of viability can be a few weeks, to hundreds of years (Germination). Germination begins when dormant seeds are under…

The extent of the denaturation of the bonded enzymes is affected by the emphasis of the pH change. In cases where the pH change is very minimal there is the possibility that the correct structure can become regenerated completely from its optimum pH becoming reached again. Ideally this procedure will result in substances that are more basic and acidic to produce less benzoquinone and substances that have a pH of seven, or close to seven, will produce a higher concentration of benzoquinone because of the preferred neutrality that a pH of seven has. When preforming this procedure one must begin with five test tubes that are occupied with the correct amount of substance that has the pH of 4, 6, 7, 8, and 10 which will be used. Once this buffer is created for each tube, 10 drops of potato juice as well as catechol oxidase containing the enzymes must be added and shaken in order to equally distribute all the components so that a possible change in pH can occur. Once placed in a 3 degree water bath the variety of colors in the substances due to the new pH change would have had a chance to completely set, making it possible to determine which pH is ideal for the most amount of benzoquinone to be…

Throughout this report you will gain information as to how temperature effects the amount of oxygen produced in an enzyme- catalase experiment. In the experiment we used liver extract as a catalase and created a chemical reaction within a reaction chamber between the catalase and hydrogen peroxide as well as three different controlled temperatures. In the procedure below there will be a step by step process as to how the experiment was created. As a result we found that the higher the temperature in the water bath the more oxygen will be produced and “rejected” from the reaction chamber. In conclusion, the reaction chamber placed in the highest temperature…
