Bacterial Unknown Report

Each student was given an unknown bacteria to figure out. I was given the unknown bacteria S38. Everybody is supposed to do all sorts of test to identify the bacteria. The first thing I did was smear my bacteria on a liquid medium. I then proceeded to incubate the medium for 24-48 hours.

1. GRAM STAIN
The next step I took in finding my unknown bacteria was to gram stain it. This is used to differentiate the bacteria. The different staining reagents are: crystal violet, grams iodine, acetone-alcohol, and grams safranin. Under the microscope it was a pink color, which means Gram Negative. Also, the shape was a rod.

    I think we correctly identified our unknown as P. aeruginosa because we performed several different tests, eleven of which helped us identify our organism on the Gram negative chart that was in our lab book. When it came down to narrowing the organisms down to ours, I looked at the glucose test first because all of the other Gram negative organisms were positive for glucose except for two. Since our organism tested negative for glucose it helped narrow it down to only those two organisms, Alcaligenes faecalis and Pseudomonas aeruginosa.     The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped. After determining the unknown was gram-negative, other bacteria’s were able to be eliminated from the options. The Triple Sugar Iron Test was chosen next because the unknown could be tested for glucose fermentation, lactose fermentation, sucrose fermentation and sulfur reduction. If the organism is able to ferment glucose and lactose and/or sucrose it will turn the medium yellow throughout (Leboffe & Burton 329). When the Triple Sugar Iron Test was complete, it showed a yellow butt, yellow slant and gas production. This determined that the unknown ferments glucose and one or both of the other sugars. The gas was produced by fermentation. The color change in the agar brought the unknown down to two options of unknowns, Escherichia coli or Hafnia alvei.     An unknown bacterium experiment, this experiment was done to discover what bacterium was in a mixed culture sample. This experiment worked with enteric bacteria which are members of the Enterobacteriaceae and live in the intestinal tract (Willey et al., 2014). Also found in Willey et al (2014) is some characteristics of enterics such as they all degrade sugars by means of the Embden-Meyerhof pathway, they produce plasmid-encoded proteins and that some of them perform mixed acid fermentation, some use enzyme systems like formate dehydrogenase and pyruvate formate-lyase (PFL) The first thing that had to be done was the streak plating to isolate the bacterium, then the next lab multiple tests were ran to help identify which bacterium was in the sample.     The unknown project was a very good realization of me and my partner going out by our selves. The very first day of the Unknown project I was acquainted with Denise who was very friendly and was very nice as far as assisting me in the project. The first day we were introduced too various different forms of the unknown such as broth, Blood agar plate, MSA plate, MAC plate, and the EMB plate. Before that we had done the 3 phase isolate which we had a possible of 10 points of achieving. In the isolation plate we had too take the sample of our unknown, which was the letter G. After we had done the 3-phase isolation plate we inoculated half of the plate, which was the S/D media. The Plate’s that we had provided too us were the BAP, MSA, MAC, and EMB. When we were successfully inoculated the S/D media the plates were put in the Incubator at approximated 37 degrees Celsius. The second day we had came into the laboratory we had too read the Nutrient Agar plate that was the one that we had too the 3 phase isolation plate. My results were somewhat correct but I had a lot of backtracking. After that we had too read the S/D Media, which we had, too diagram and describe each of the plates that we had put in the incubator. Once they had come out of the Incubator the results that I had achieved was the MSA plate was yellow, stinky, I did have growth, there were a positive match for small colonies, and was shiny. The second one that I had achieved was on the MAC plate. The color on this specific plate was red, it had smelled rotten, there was definitely some growth on the plate the texture was shiny and was not raised. Out of these plates I had somewhat of a indication of what the specimen was since after the gram stain we had done in lab we found out that on the MSA plate we had achieved Gram positive…

    The gram staining procedure consists of fixing a colony of bacteria onto a slide and then flooding the colony with various chemicals. First, crystal violet dye was dropped onto the bacterial cells, staining gram-positive cells purple. Iodine was added to fix the violet dye into place and then ethanol was used to wash the dye off the unstained cells. Finally, a red dye called safranin was used to stain any gram-negative cells present so that they were visible. Once the gram stain procedure was complete, the gram-positive bacteria appeared purple under a microscope while gram-negative cells appear pink or red. The purpose of this lab was to differentiate bacteria based on the cell wall and to differentiate whether the bacteria was gram-positive or gram-negative.
