You have been provided a mixed culture (a gram positive and a gram negative) from which you should have isolated each type of organism, described their colonies on a TSA plate, and described their cellular morphology. You were graded on this activity separately. After you have isolated your gram negative in pure culture you will be subjecting it to various biochemical tests and making observations about the cells, colonies, growth temperature preferences, etc. in order to identify it. You will provide a report (no more than 4 single-spaced pages including the dichotomous key tree and table) on your identification that includes the following:
|1. |An identification tree. You must key out your organism from all others listed in the unknown list (available in the Blackboard course site). When a |
| |particular organism has been eliminated from your list due to a test result that does not agree with your own, provide the name of that organism under that |
| |test result on the tree (spell correctly, italicize!). |
| |Note that, in general, an organism name appears ONCE on your tree—at the point it was eliminated as a choice. The path to your identified organism should be |
| |highlighted. This should be typed. Word and Powerpoint are both programs that will allow you to work with flow charts that can be constructed as a tree. If |
| |in Powerpoint, change the slide to a custom size that is 8.5 x 11 inches (a page). You can work in text boxes that can be dragged around and connected by |
| |lines later. Instruction hints for a simple tree in Word will be uploaded as well. Collaborate with more tech-savy classmates if you are having trouble. Once |
| |you know the right buttons to click it’s not difficult. Typed, neat, correctly executed = 12 points
References: (include only those references that have been cited somewhere in the body of your report. Use appropriate format. Noodlebib is recommended (on | | |library website) | | | Correctly executed = 7 pts | This is a mini-investigation and formal report. Take it seriously as a poorly executed report will reflect negatively on your grade regardless of the identification made. The report must be type written, including the trees. Please be aware of spelling, sentence structures, and grammar. Always capitalize genus and use only lowercase for species and italicize them. After the first use you may abbreviate the genus (E.G. 1st mention= Escherichia coli ( E. coli after it was spelled out once) This report will take work but we will work on the data collection for it over several weeks. Good notes are going to be key for your report construction! A well done report ALSO reviews a lot of important material that may appear on the lab final (
