Title: The effect of the concentration of hydrogen peroxide on catalase
Aim: to examine how the concentration of the hydrogen peroxide affects the rate of reaction
Hypothesis: The substrate concentration will determine the rate of reaction. The reaction will increase as the concentration increases, but from a certain concentration, the rate will maintain the same rate
Materials:
3% hydrogen peroxide
6% hydrogen peroxide catalase round metal basin measuring cylinder thermometer conical flask rubber stopper and delivery tube
50mL beaker pipettes blue tack stopwatch Method:
1. Refer to apparatus below for set up

2. Pour 10 of 3% hydrogen peroxide into measuring cylinder and pour into beaker. Add 0ml of water to the beaker.
3. Pour substance from beaker into the conical flask
4. Attach rubber stopper with delivery tube to the neck of conical flask
5. Place 2mL of catalase in the pipette and release catalase though rubber stopper. Immediately place blue tack on the opening and start timer.
6. Observe the reaction. Stop timer once water level reaches 20mL
7. Record results
8. Repeat experiment from 2-7 with hydrogen peroxide at 8mL with 2mL of water, 6mL hydrogen peroxide with 4mL of water, 4mL hydrogen peroxide with 6mL water and at 2mL hydrogen peroxide with 8mL water

Risk Assessment:
In this experiment, hydrogen peroxide can be very dangerous and toxic if inhaled or has contact with skin or eyes.
Therefore wearing safety goggles while handling with hydrogen peroxide. Tying up hair and removing any loose clothing such as ties, blazers in which may come into contact with the hydrogen peroxide.
Spillages should also be cleaned up immediately to avoid any accidents and contact of substance to skin.

Purpose: Cells produce toxic wastes, in this experiment hydrogen peroxide, and without some sort of molecule to break it down the cell will die, along with the organism itself. However with the aid of an enzyme, catalase, hydrogen peroxide is able to be broken down into an intermediate and thereafter harmless substances water and oxygen. The goal of this lab is to measure the reaction rate of this process in different substances such as a liver, a vegetable, and breast tissue. By using variables such a pH and temperature we are able to how the rate of reaction is altered or improved. If it has improved, the optimum has been discovered and the enzyme will create a higher reaction rate. If above the optimal points, proteins will denature and the reaction rate will remain the same. This is vital for cellular activity for if homeostasis is not reached enzymatic activity will decrease or the enzymes will simply denature and the toxicity within the cell will increase killing the cell.…

Next, 50ml of distilled water will be measured using a graduated cylinder. The 50g of catalase and the 50ml of distilled water will be put into a blender and blended until liquefied. The catalase solution will then be introduced to a Buchner funnel filtration system to separate the catalase extract from any solid particles left over from the blending process. This step will be repeated until all of the catalases have been blended and filtered. Next, 5 ml of hydrogen peroxide will be measured out using a graduated cylinder. It will then be poured into a small Erlenmeyer flask. This will be repeated five times, representing the five trials for each catalase treatment. Once the Erlenmeyer flask has been given 5ml of hydrogen peroxide, 10ml of the S. oleracea catalase will be measured out and poured into the Hydrogen peroxide filled flask. When the catalase and Hydrogen peroxide meet a reaction will occur and the hydrogen peroxide will be broken down into water and oxygen. The oxygen released from the reaction will be recorded by the Gas Pressure sensor. This step will be repeated five times for each necessary trial, then the next catalase extract will be…

The enzyme peroxidase has been shown to break down H2O2. Enzymes are known to increase the rate at which a chemical reaction occurs. We looked at factors that affected the breakdown of hydrogen peroxide. These effects are the different temperatures and pH levels the enzymes were placed in. We found that the optimum, or best condition, temperature for the enzymes tested was about 22 degrees Celsius. The optimum pH level for the enzyme was 7.…

First, we used a clean syringe labeled H2O2 and filled it with H202. Then, we transferred the contents of the syringe into a 60 mL cup labeled Baseline. Second, we used the plastic transfer pipet to add 1 mL of distilled water and added it to the Baseline cup. Third, we used the syringe labeled H2O2 to add 10 mL of H2O2 and transfer that into the Baseline cup. Fourth, we gently swirled the contents of the Baseline cup to mix the solution. Then, we used the syringe labeled Transfer and removed 5 mL of the solution in the Baseline cup into the cup labeled Titration. Lastly, we titrated the 5 mL sample of the Baseline solution.…

This investigation examined what would happen to the rate of an enzyme-catalyzed reaction if the concentration of substrate changed. We hypothesized that if the concentration increased, then the reaction rate would also increase. To test our question, we varied a combination of substrate and buffer, totaling 6mL, with a constant amount of 2 drops of catalyst. The enzyme catalyst, peroxidase, increased the rate of the reaction. The results of our experiment can be found by comparing the reaction rates for each trial. These rates are actually the slopes of the lines that were graphed during each reaction. The trend of data…
