The Introduction

A Biomolecule is any molecule that is in the human body. There are four major biomolecules, they are Carbohydrates, Lipids, Proteins, and Nucleic Acids and they all have different functions and characteristics to classify an organism. Carbohydrates are the most important source of energy for your body, this is mainly sugar. Lipids are molecules that contain hydrocarbons and make up the structure and function of living cells. Nucleic acids is an organic system present in living cells, especially in DNA or RNA. A protein is very essential for the living body its is made up of amino acids. In each station there was a positive and a negative control. If the substance was the same as the negative control then it was negative. If the substance was the same as the positive control than the substance is positive. In the first lab we did the Paper Spot test, one would have had to apply the substance on a cardboard like paper. If it looked oily it was the biomolecule called lipid. The result of test one was it stayed the same, it did not turn oily and was clear. Next was the test called Biuret test, this was to test if it was a protein. If it was a positive control it would break down into 2 colors of maroon-brownish. While substance 1 stayed clear with a blue-green tint. Concluding that the substance is definitely not a protein. The next test was Benedict's Test this was to show if it was a carbs-sugar. One had to apply a substance with another substance once mixed one had to have it boil for about 5 minutes. If it was positive it would turn into an orange clear liquid.The substance after the 5 minutes turned into an orangish liquid this concluding that it was a carb-sugar. Lastly is the Iodine test, this test was show if it was a carbs-starch. Then the substance had to mix the mystery substance with iodine, if positive it would turn into a blueish-black. As a result the substance changed into the color and was also positive for carbs-starch.

    A few experiments were performed to test for the presence of the different types of macromolecules of various solutions. The iodine tested for starch and glycogen, Benedict’s test tested for reducing sugars for carbohydrates and biuret test was performed to test for proteins. It is necessary to understand how important controls are in a lab experiment. A control is where all of the experimental variables can be controlled and monitored. Results are determined by comparing these variables to the control. Without them, no results would be valid in the experiment.     If the samples did contain the antigen then the primary and secondary antigen would react and bind to the antigen and the enzyme substrate, therefore resulting in a blue color change. It is important to wash out the wells after every step, to keep the experiment all the same, and to make sure there is no unwanted substances inside the wells, that would interfere with the experiment. When we added the primary antibody to the well, and it contained the antigen, then the primary antibody would bind to the antigen, and if the sample did not contain the antigen, then the primary antibody could not bind to the antigen. But in both these situations, there would be no color changing reaction, because the secondary antibody was added and couldn't bind to the enzyme substrate, which causes the blue color changing reaction if its positive, and no reaction if its negative. If the sample shows a negative result this does not always mean you no not have the disease. This could always be because the experiment wasn't done properly and there was an error somewhere along the process which caused the result to appear negative, or that the sample taken did not contain any of the disease or the antigens, and it could still be some of the disease somewhere in the body.     Learning experience 4 teaches the understanding of how to identify a small sample of Biomolecules within a wide variety of substances. The specific biomolecules that were observed in the lab were Sugar, Starch, Protein, and Lipids. These 4 Biomolecules were tested within the foods applesauce, peas, and turkey gravy. These biomolecules were identified using the solution indicators for the Biomolecules which are respectively designated for each biomolecule. Iodine for Starch, Benedicts for Sugar, Biurets for Protein, and a brown paper bag for Lipids. The biomolecules were observed when the food and indicators were combined, the solution then turned a color designating a specific biomolecule. Protein turns purple, Iodine turns black/blue, Sugar turns orange, and since lipids was actually not mixed with another solution and instead dropped onto a brown paper bag, would reveal a grease stain over time to indicate whether or not the substance contained Lipids. Two other major biomolecules that were not tested in the lab were Nucleic Acids and Carbohydrates. If we study the foods before the lab we can observe what biomolecules may be in the foods. Since Turkey Gravy contains meat, it may contain Protein. Since Applesauce comes from an apple, it should contain some sort of natural sugar.
