This enzyme is required for the synthesis of prostaglandins which regulate hormones, inflammation, and calcium movement. This creates the analgesic, and anti-inflammatory properties
Pharmacokinetics:
Absorption: "When absorbing, the peak plasma levels of celecoxib occur approximately 3 hrs after an oral dose. Under fasting conditions, both peak plasma levels and area under the curve are roughly dose-proportional up to 200 mg twice daily; at higher doses there are less than proportional increases in plasmas levels and the curve." Distribution: Celebrex is a highly bound protein, 97% of it is bound to plasma proteins. It primarily binds to albumin and a little lesser to glycoproteins. This resul!,in a extensive tissue distribution at a consistent rate.
Metabolism and Excretion: Mostly metabolized by the hepatic CYP2C9 isoenzyme; the CYP2C9 enzyme system exhibits genetic polymorphism. The other two are carboxylic acid and its glucuronide conjugate. The half life of Celebrex is about 11 hours. Once at mination it has been found to mainly be excreted through the feces with a small amount of the unused drug excreted through the urine. 7 0s- t.C..7
