Gatorade Spectrophotometer
The purpose of this lab was to use a spectrophotometer to find the concentrations of glucose in varying samples of Gatorade. The varying executions included changing the temperature of reaction (room temperature instead of 40 degrees centigrade), changing the time for reaction (20 minutes in the water instead of 45 minutes), and changing the amount of enzyme added (0.1mL of enzyme. The results in from this lab gave data points with respect to ferricyanide. Fortunately, ferricyancide and glucose have a 1:1 ratio meaning that the amount of glucose can be deduced. For the first 7 samples required the spectrophotometer to determine the absorptivity, which was run at 420nm. These data points could then be used to find a line of best fit for the later calculations. The R^2 value was given at 0.986. This was after deleting a value from the data set because it was pulling the R^2 value down. The line of best …show more content…
One source of error came from washing out the test tube with the next sample before running the test. This means that there could have been remnants of the old sample in the new sample. Another source of error could have been from lack of precise timing causing the reaction to not go as long or longer than desired. Of course, each solution was made by hand leaving room for human error. These errors include over injecting of solution.

Conclusion
This experiment found the amount of ferricyanide which was then used to find the amount of glucose thanks to a 1:1 ratio. This could then be used to find the concentration of glucose in various samples of Gatorade (15 solutions). Then the lab was split altering 3 different variables (Time, Temperature, Enzyme amount). This illustrated the variable that had the biggest impact on the results.

LAB QUESTIONS
1. No, it would not work because sucrose is a polysaccharides but this enzyme interacts with monosaccharides. This could only work if they were broken down or if a different enzyme was

Glucose exposure resulted in a yellow color change proving that Unknown #7 fermented glucose and produced an acid but when exposed to Sucrose Unknown #7 produced a red color change indicating that it was a non-fermented of sucrose which was reflected with an alkaline bi-product. The Enterotubes showed that Unknown #7 was an "enteric" bacterium with a score of "46141" Lysine, Adonitol, Lactose, Dulcitol/Phenylalanine, and Harnstoff/Uree were all negative, while Ornithine, Arabinose, and Sorbitol were proven to be positive. The IMViC test identifies members of Enterobacteriaceae and included Indole that tested negative, Methyl Red which produced a red color change showing that Unknown #7 could ferment glucose to acidic ends, Vogves Proskaer which tested negative because Unknown #7 failed to convert those acidic bi-products back to neutral ends, and Citrate which tested positive with a blue color change and growth on the medium because Unknown #7 was able utilize citrate. The Nitrate Reduction test displayed Incomplete Reduction with a red color change when Solution A and B were added meaning that it could only reduce NO3- to NO2-…

The Fehling’s test was done to show the presence of carbohydrates. Two small test tubes were used and 5 mL of Fehling’s reagent was added to it. Five drops of 1% glucose was added to one test tube, the control. This tube was then boiled and if a brown-red precipitate was formed, it would indicate that there was a carbohydrate present. The other tube had 5 mL of the Fehling’s reagent put in it and five drops of the filtrate after the ethyl acetate was gone and the filtrate had been cooled. If this created a brown-red precipitate, it would indicate that there was a carbohydrate…

In this lab there were many sources of error that could have affect our experiment. One of this possible errors could have been using a wet or a beaker with another substance inside of it that would have other compounds or residue from other past experiments. This source of error would cause the ions in the solution to dissolve with different compounds and the percent yield would be affected by that. Additionally, leaving some water left behind in the solid product which would leave ions in the solid product and this would ultimately mess up test 1 and test 2 by reacting the wrong…

Sucrose was successfully transformed into D-glucose and D- fructose (disaccharide to monosaccharides) through acid-catalyzed hydrolysis. This is obvious when comparing the experimental specific rotations of sucrose and invert sugar with the theoretical values of sucrose (+66.5°) and invert sugar (-39°). However, my partner and I experienced a small complication with the addition of the acid, HCl. After adding the required amount of HCl to the mixture, we experienced no color change for about an hour. Due to this problem, we came up with a solution to add more HCl due to the possibility that the acid was absent from the reaction. An additional 16 drops of HCl was added during the heat reflux phase. The solution displayed a tint of yellow after about 5 minutes of refluxing, and then it was completely yellow after another 5 minutes. Although we added additional amounts of HCl, it is not possible to determine if the inverted sugar contains an excess amount of HCl. The inverted sugar refluxed for a total of one hour and 10 minutes due to a restricted amount of time to complete the…

It is often important to measure the concentration of glucose in a solution. In this experiment a variety of solutions will be tested for the glucose concentration of known value and a graph, drawn to show the results. This type of graph is known as a Standard Curve. This graph will then be used to estimate the glucose concentration in an unknown solution. This is the method which was used in hospital labs to measure the glucose level in blood samples. Glucose (C6H12O6) is a monosaccharide reducing sugar. In this reaction the glucose readily donates electrons which are accepted by the permanganate causing it to change colour. The time taken for the pink colour of the potassium permanganate to disappear…

Results: The data supported the hypothesis that the 15 gram fructose solution had the greatest production of CO2. This is shown by the steady rate of respiration whereas the others showed less change or minutes at a time where there was no change in mm of CO2 produced. Also, as shown in the table, the vial with the greatest fermentation rate after solving measurements was the 15 gram fructose solution. As expected, the control vial containing only yeast showed no carbon dioxide production because there was no sugar to break down during…

The spectrophotometer was plugged in and heated up for at least 15 minutes. The blank test tube (containing all components except iron solution) was then added to the spectrophotometer and set to 0 absorbance/100% transmittance. The absorbance of each solution was measured at 510 nm quickly after they were mixed. The table on the next page shows the absorbance of each test tube along with the micrograms of iron they contain.…

A spectrophotometer or colorimeter makes use of the transmission of light through a solution to determine the concentration of a solute within the solution. A spectrophotometer differs from a colorimeter in the manner in which light is separated into its component wavelengths. A spectrophotometer uses a prism to separate light and a colorimeter uses filters.…
