Glioma Essay
Malignant astrocytomas or Gliomas are the most frequent form of primary cancer that arise in the brain. The most malignant and aggressive type of glioma is glioblastoma multiforme(GBM). GBM ,which called grade IV astrocytoma, is one of the most common and deadly primary malignant brain tumor in adults with a high degree of morbidity and mortality and accounts for about 15% of all intracranial tumors in adults ages 40–75(2, 3)
Incidence rates of glioma in The United States yearly is about 5 in 100,000 persons (4)
Despite progress in chemotherapy, radiation oncology and surgical the prognosis of this cancer remains extremely poor (7-9)
On the other hand, Their anatomical localization, the penetration of the enviroment normal brain parenchyma

Among other pathways, The TGF-β pathway has been implicated to play a key function in glioma initiation and progression due to its effects on cell proliferation (20), tumor invasion (21), angiogenesis (22), immunosuppression (23) and the maintenance of stemness of glioma stem cells (GSCs) (24).
TGF-b has also been reported to induce matrix metalloproteinase (MMPs) expression and suppress tissue inhibitor of metalloproteinase (TIMP) expression, and increase the invasion of human glioma cells(19, 29) ligand-bound type II receptor activates and phosphorylates the type I receptor. Subsequently, Activated TbRI initiates cytoplasmic signaling pathways, which then phosphorylates Smad family proteins, Smad2 and Smad3 in the TGF-b subfamily, eventually that can pair with Smad4, a family member that is not phosphorylated by the type I receptor. also, The receptor complex recruits adaptor proteins and via post-translational modifications and activates several signaling proteins that transmit biological information (30,

Glioma is the most common primary brain tumor [1, 2] and it is one of the most rapidly growing, therapy resistant and devastating cancers [3]. High-grade glioma patient’s prognosis is very poor [4, 5] and the median survival is only 14.6 months after receiving all the currently available treatments [1]. A new effective therapeutic approach is urgently needed to give some hope to the glioma patients.…

Target volume definition for re-irradiation (Re-RT) of recurrent glioma generally includes any contrast enhancing lesions on T1-weighted MRI, adding a safety margin of approximately 5mm [5]. To encompass the real dimension of the tumor, the additional benefit of amino acid PET has been evaluated and is currently being assessed in a prospective clinical trial. On important factor on the outcome is extensive neurosurgical resection of glioma recurrences, where several groups have shown that the extent of resection (EOR) is associated with outcome [13-15]. However, in alleged completely resected glioma recurrences, Re-RT is withheld – most likely due to historical reasons.…

    Gleason Stereotypes

Several methods for grouping of Gleason scores in prognostic categories have been proposed. Gleason scores 2 to 4 behave similarly and can be grouped. Gleason scores 8 to 10 are usually grouped together as they represent poorly differentiated tumors. Gleason score 7 is a distinct entity with prognosis intermediate between that of Gleason scores 5-6 and 8 to 10, respectively.21Hence we grouped the patients into four groups wherein Gleason score of 2-4 represent well differentiated tumors, score 5&6 intermediate grade tumors, score 7 moderately differentiated tumors and score 8 and above high grade tumors.…

While before there was no permanent cure for this common cancer, after this new treatment, the outcomes are promising. Normal life without all the effects of the tumor or the effects that come along with chemo or radiation therapy lead to healthier overall patients. With further procedures and testing, there is now a light at the end of the tunnel for the many who suffer from Glioblastoma…

GDF15/MIC-1 is a product of macrophage activation, suggesting that it may be involved in chronic inflammatory processes (-). GDF15 is a stress-responsive member of the TGF-β superfamily and initially described as macrophage inhibitory cytokine (-). Although GDF15 has been described in many solid tumors for almost 10 years, the nature of its receptor is still unknown. There is some evidence of Smad signaling pathway activation, suggesting GDF15 may act through TGF-β receptor superfamily pathways (-). The TGFβ family belongs to a group of cytokines with pleiotropic functions (-). This family has 33 members, including growth differentiation factors (GDFs) and TGFβ. They are involved in the regulation of various biological functions such as proliferation, differentiation, migration, and apoptosis in many different cell types (-).…

It is well known that Gliomas are amongst the most destructive types of cancers out there. Unfortunately, Gliomas are the most frequent tumors in the central nervous system accounting “for more than 32% of all primary brain and CNS tumors and 80% of all malignancies of the brain and CNS”. Tons of research has been done in this topic with a variety of different methods, however, it still has a poor prognosis with frequent recurrences. The gold standard so far to treat this type of tumors has been surgery, radiotherapy, and chemotherapy. However, a huge dilemma many practitioners face while treating this type of tumors is the blood brain barrier that acts a shield for this tumors and restricts the delivery of therapeutic molecules into the brain.…

The fundamental understanding of Glioblastomas consist of knowing the structure, origin, and symptoms of this type of cancer. “According to the American Brain Tumor association Glioblastoma has been described as “tumors that arise from astrocytes—the star-shaped cells that make up the “glue-like,” or supportive tissue of the brain. These tumors are usually highly malignant (cancerous) because the cells reproduce quickly and they are supported by a large network of blood vessels.” In addition, this form of cancer attacks surrounding tissue which prohibits the ability to spread across the body irregularly. GBM tumors are generally found in the cerebral hemispheres of the…

My aunt and I were very close throughout my whole life, but she was diagnosed later in her life with pancreatic cancer. I was affected by this because of how close we were and how much time I had spent with her; she was like a second mother to me. Losing her to this cancer was devastating and really hard on our whole family. Ever since her death in 2010, I had been curious about this pancreatic cancer. Researching this topic is very important to me because I want to learn more about this cancer and get a better understanding of what goes on when one who is diagnosed. This paper will not only educate myself about the cancer, but will educate others as well.…

    73. They combined the proliferation-invasion model with information on the molecular level dynamics of tumor growth referred to as the proliferation-invasion-hypoxia–necrosis–angiogenesis (PIHNA) model. Their model characterized glioblastoma tumor by partitioning the malignant tumor cells into subpopulations including normoxic and hypoxic cells, necrotic tissue, and neo-angiogenic vasculature and angiogenic factors. Using functional imaging, they evaluated the biological interaction of metabolic changes within and between cells on a patient-specific level and suggested that changes in cell kinetics are not essential to generate the imaging and histologic features of malignant progression and rather interactions of tumor cells with a constant invasion and proliferation rate alone play a key role in malignant progression. They compared survival map for different grades of glioma with the grade map and concluded that the shortest survival times are for those tumors with both high D and ρ, in contrast to WHO grading scheme which suggests that only high ρ is important. An important area of future…

The introduction of the paper gave sufficient basic information on Glioblastoma multiforme. It was stated that GBM cells have rates of high proliferation within the brain. The astrocyctic tumors express many integrins which promote specific inhibitors and growth factors. When the PKCs are upstream along with other growth factors they aid in the progression of differentiated astrocytic tumors. The materials and methods gave information that best suited the goal of the paper. Methods consisted of cell cultures, retroviral expression, propidium iodide FACS analysis, H-thymidine incorporation, and western blot analysis. The only section not fully understood was the use of the H-thymidine method. The data and graphs set up were helpful to understanding the results. This was observed throughout each figure given. Figure 3, 4, and 5 especially shows the presence and lack of activity of all samples in different…
