Gram Staining is a technique used to separate two groups of bacteria based on their cell walls. The two groups are gram positive or gram negative.
2. What is the purpose?
The purpose of gram staining is to determine if a bacterium is gram negative or positive. If it is gram positive that means the stain will be purple and that the bacteria have a thick cell wall made of peptidoglycan. Gram negative stains pink and can cause infections like pneumonia.
3. What are the necessary steps?
• Apply a primary stain (crystal violet) to a heat fixed smear.
• Wash excess stain with water and add a few drops of iodine solution on the smear for 1 minute.
• Rinse in water and wash with 95% ethanol for a minute
• Rinse with water then stain with 1% safranin for a minute.

This test is very beneficial because if a person presented to their pcp he would order a gram stain to learn the type of bacteria that is causing your illness and based on the result he would determine the proper course of
