Observing Bacteria and Blood

Questions:

A. List the following parts of the microscope and describe the function of each

A. Eyepiece lens B. Tube C. Revolving nosepiece/turret D. Objective lenses E. Stage F. Iris Diaphragm G. Illuminator H. Coarse Focus I. Fine Focus J. Arm K. Stage Clips

B. Define the following microscopy terms:
a. Focus: A means of moving the specimen closer or further away from the objective lens to render a sharp image.
b. Resolution: The ability of a lens system to show fine details of the object being observed.
c. Contrast: A circular plate placed on the stage of a low power microscope. One side is white, the other is black. It can be flipped around depending on the coloration of your specimen.

C. Describe your observations from fresh yogurt slide you prepared in Part III. Several cells majority Bacillus and cocci cells. D. Were there observable differences between your fresh yogurt slide and the prepared yogurt slide? If so, describe them. Only perhaps that there was far more Cocci cells then Bacillus

E. Describe the four main bacterial shapes

1. Cocci - Spherical or oval bacteria having one of several distinct arrangements based on their planes of division
2. Bacillus - Rod shaped bacteria. Bacilli all divide in one plane producing a bacillus, streptobacillus, or coccobacillus arrangement.
3. Spirillium – Rod shaped bacteria that appear thin and spiral shapped.
4. Vibros - Oblong shape that has a similar shape to peanut shells in that their curvature.

F. What are the common arrangements bacteria are found in? Cocci, Diplococci, streptococci, staphylococci, bacillus, diplobacillus, streptobacillus.

G. Were you able to identify specific bacterial morphologies on either yogurt slide? If so, which types? I believe I could see cocci, and diplobacillus.

H. Describe the cells you were able to see in the blood smear. Red blood

Specific bacterial morphologies noted in exercise one come from the spiral bacteria sample. The shape appears to be corkscrew in nature and vary in length throughout the sample slide. The length varies but the width seems to be uniform. The color most likely arises from the stain used to allow for better visualization of the organism. The Bacillus sample has no discernable morphology and appears as round ended, and cylindrical shaped in nature. The organisms have a black tint which is probably related to the type of statin used for visualization of the organism.…

The tests performed on the unknown bacteria cultures were all used to determine the identity of the bacteria. Each of the tests performed provided some key information about the bacteria in question and how it functions. Not all of the tests were performed on every culture, however, as some of the tests were used only for gram (+) or gram (–) bacteria, while others were even more specific and used only for cocci bacteria. The tests performed and what constitutes a positive and negative test are as follows.…

Introduction: There is bacteria that is beneficial and harmful in the human. Yogurt was choosed to be the best example that contains this bacteria. What happens to be the difference between these two types of bacteria, and how are they harmful? The bacteria is considered “Gram-Negative,” and the substance is considered contaminated when there is more negative bacteria than positive bacteria. Bacteria are very different from viruses. First of all, bacteria are much larger in size. The largest virus is only as big as the very smallest bacterium (singular for bacteria). But bacteria are still microscopic and cannot be seen with the naked eye. They are so small that the sizes of bacteria are measured in micrometers (10,000 micrometers = 1 centimeter). By comparison, the head of a pin is about 1000 micrometers wide. Though more complex than a virus, the structure of a bacterium is still relatively simple.…
