Experimental Methods
In this lab, we use the procedure from Chemically Defined, Complex, Selective, and Differential Media. We divide the agar plates that have the unknown in third, write our initials, and date on the bottom of the agar plate. We also divide the agar plates into quadrants, label each organism in their quadrant, write our initials, and date on the bottom of the agar plates. It appears to be differential media because it has colonies growing on the agar plate. The appearance of the organism looks different from each other and we can take those colonies, and isolate them from the other organisms. EMB is the purple color plate and each organism within the quadrant has different colors. We streak the S. epidermidis quadrant but it has no color. E. coil on the EMB appears metallic green and P. aeruginosa appears dark purple on the agar. E. coli turns green because of the sugar lactose. EMB is differential media because of the green color of E. coil and it also selects for gram negative and inhibits gram positive. TSA plate is clear and has white color. E. coli grows best on Mannitol plate, E. averogenes grows best on the EMB, and finally E.coli also grows best on the TSA as well. The results for the known organisms are fantastic and they have a lot of growth. The TSA below has a great combination of colors such as yellow and cream white color, but some TSA have green color as well. It is kind of hard to see the yellow color in the picture. The image looks like a bowl of soup because of the appearance. E.coli on the EMB plate appears green and it is selective for gram negative, and inhibits gram positive. Overall, the known organisms depict beautiful colors. The unknown number 5 is inclusive. Klebsiella pneumoniae might grow well on EMB because of the gram negative lactose fermenter, and it may selects gram negative and inhibits gram positive. The organism may have E.coli appearance to on the TSA and BHI may not like this organism since BHI is very picky organisms.
