Neuromodulators have been key elements in all neurological functions in the central nervous system (CNS). Those chemicals are released from a neuron affecting other groups of neurons in the CNS. Serotonin (5-HT) is a crucial neuromodulator that modulates synaptic efficacy in the nervous system. It is most well-known for its role in the brain where it plays a major part in mood, anxiety and happiness (1), it therefore, strongly affects our social life. In the nematode Caenorhabditis elegans, many behaviors are regulated by 5-HT levels, which are in turn regulated by the presence or absence of food (2). Behavioral assays will be done on the model organism, C. elegans, to see their responses to the different stimuli, and their effects on 5-HT levels. …show more content…
ASH neurons are special polymodal nociceptive sensory neurons found in Caenorhabditis elegans. Special interest is dedicated to those neurons because they are the key cellular elements responsible for avoidance behaviors in response to chemical repellents, high osmotic solutions and nose touch (3). My goal is to further and better understand neuromodulator’s effects in the nervous system. Which will help us understand the regulation of diverse populations of neurons in our brain.     The body of Caenorhabditis elegans (C. elegans) are surrounded by a cuticle that is molted four times in development although not all cuticle collagen genes are expressed at once. Mutations in these genes lead to morphological defects such as blister formation in the cuticle. The blister phenotype (Bli) is inherited recessively and develop in the adult stage of C. elegans. Other cuticle collagen mutations, such as the uncoordinated (unc), roller (rol) and dumpy (dpy) mutations, can suppress the Bli phenotype. Sc109 is also another mutation that is found in a carrier of the bli-1 allele and was found to incompletely suppress the formation of blisters. Tetraspanin protein (TSP-15) is found to be needed to maintain the integrity of the epithelial membrane as a deficiency of TSP-15 lead to separation of cuticle layers. Dpy-5 alleles are dominant suppressors of bli-4 mutations although suppression is incomplete.     Serotonin has a range of influences on the neurological and physiological function of the body. It has a significant influence on sensitivity to pain, emotionality, and a behavioral response to positive and negative consequences. Serotonin, along with other major neurotransmitters, is also linked with psychotic disorders such as schizophrenia. Serotonin, or HT-5, also has an effect on sleep, eating patterns, and thermoregulation (Meyer et al., 2005). These seem to contribute to depressive symptoms in animals with depleted stores of 5-H. There are now 14 discernable types of serotonin receptor subtypes. Subtypes 2B, 2C, 4, and 6 have all been linked with depression and anxiety (Tohda et al., 2006). Recent research shows that altering 5-HT2CR mRNA to track the origin of depressive diseases may reveal conclusive information about the origin of neurochemical imbalances. (Tohda et al., 2006) There are different approaches to raise serotonin levels in the brain to normal levels. The most commonly prescribed antidepressants are classified as selective serotonin reuptake inhibitors or SSRIs. SSRIs regulate the serotonin transporter, SERT, which slows the reuptake of the neurotransmitter into the presynpatic terminals. While new, more effective drugs with fewer side effects are being deliberated there have been newer ways to attempt to regulate serotonin imbalances. A recent discovery of small interfering RNA molecules may drastically change the treatment of depression and all other neurological diseases.
