Ornithine Decarboxylase Test Report
The Results from the Gram positive tests indicated that the unknown #4 was Streptococcus pyogenes. All seven tests on the unknown matched S. pyogenes perfectly. The blood agar plate proved the unknown to be β hemolytic, meaning the unknown bacteria was capable of complete hemolysis. This test separated the unknown into the β Streptococcus group, narrowing the possible bacteria to S. aureus or S. pyogenes. The Catalase test was used to determine if the unknown could break down hydrogen peroxide. The unknown bacteria was unable to break down the hydrogen peroxide, meaning it was catalase-negative. This result from this test correlates with the S. pyogenes and is in contradiction with S. aureus, a strong indication that the bacteria is in fact S. pyogenes. The results show that the unknown and E. aerogenes were positive for this test while P. aeruginosa was negative. Results for P. vulgaris were negative for this test, but this bacteria can show positive or negative for the Ornithine, The lactose fermentation test (MAC) showed that the unknown was positive for lactose fermentation. Expected results showed that all bacteria should have been positive for this test. However, the test on P. vulgaris during the experiment was negative, defying expected results. The oxidase test was used to test for the presence of cytochrome c in the unknown. The unknown was shown to be oxidase negative which matches with the results for E. aerogenes. The results for P. aeruginosa were positive, making this bacteria less likely to be the unknown. The results for P. vulgaris were negative, matching expectations for this bacteria. The final test was the SIM agar test. The unknown was negative for motility, indole, and H2S.     I think we correctly identified our unknown as P. aeruginosa because we performed several different tests, eleven of which helped us identify our organism on the Gram negative chart that was in our lab book. When it came down to narrowing the organisms down to ours, I looked at the glucose test first because all of the other Gram negative organisms were positive for glucose except for two. Since our organism tested negative for glucose it helped narrow it down to only those two organisms, Alcaligenes faecalis and Pseudomonas aeruginosa.     The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped. The Triple Sugar Iron Test was chosen next because the unknown could be tested for glucose fermentation, lactose fermentation, sucrose fermentation and sulfur reduction. If the organism is able to ferment glucose and lactose and/or sucrose it will turn the medium yellow throughout (Leboffe & Burton 329).
