Unknown Microorganism. 17 Lab Report
In order to properly performed this test, the initially slant for unknown microorganism #17 is important. For this lab, two identical slants are used for two reasons. Firstly, the slant can be used to make sure that there is no contamination from the Nutrient Agar plate. Secondly, the second slant will become a stock culture to prevent the shortage of slants during performing the series of tests.
Kliger’s Iron Agar tests can be used to determine multiple characteristics of unknown microorganism #17. Kliger’s Iron Agar slants also contains a pH indicator, phenol red, which can be used to test the presence of fermentation. If there is a glucose or lactose fermentation, the acid will be produced, and the color will change from red to yellow. If there is only glucose fermentation, the slant will have a yellow butt. If there are both glucose and lactose fermentation, there will be a yellow slant and butt. Kliger’s Iron Agar slants is also used to check the presence of hydrogen sulfide. If hydrogen sulfide reacts with ferrous sulfate, the slant will have a black butt, and the test result is positive. Otherwise, if the slant has a yellow butt, the test result is negative. This agar also contains a pH indicator, bromothymol blue, which can be used to identify the presence of oxaloacetate and pyruvate (1). If the slant changes to color blue, which indicates that the microorganism is able to produce citrate, and the test result is positive. Otherwise, if the slant remains a green color, which indicates that the microorganism is not able to produce citrate, and the test result is negative.     The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped.
