You were given five samples of patients’ urine and some Uristix® dipstick. Describe how you used the dipsticks to determine if urine sample contained amounts of protein and / or glucose.

To test the given samples for glucose or protein, dipsticks were immersed into each of the urine samples for 60 seconds. The colour change was compared with the colour chart on the Uristix® bottle.

The detection of glucose in urine is based on the presence of two enzymes, glucose oxidase and peroxidase. The first enzyme, glucose oxidase, catalyses the formation of gluconic acid and hydrogen peroxide from the oxidation of glucose. The second enzyme, peroxidase, catalyses the

It was the only tube that contained just BSA, without any diluent. The second tube was prepared by adding a 250 μl BSA and 250 μl of 0.9 % Saline (diluent). Following two tubes were prepared with the same amount of diluent. However, the volume of BSA added was different. Therefore, 250 μl from the tube 2 was pipetted to the tube 3, then the same amount was pipetted from tube 3 to tube 4. The fifth tube contained 800 μl of diluent and 200 μl of liquid transferred from tube 4. Lastly, tube 6 was a ‘blank tube’, containing only 250 μl of diluent.     The homogenates provided were made by homogenizing tissues in a sucrose phosphate buffer in a 1:20 ratio. The protein concentration in bovine cells was measured by diluting the homogenate with a 1:5 ratio; 50 microliters of homogenate and 200 microliters of water. Then 5 known protein concentration samples which were 0.4, 0.8, 1.2, 1.6, 2.0 mg/ml of bovine serum were used to determine absorbance with a spectrophotometer. Two additional samples were made; one was blank and the other was for the specific homogenate sample. Then 3 microliters of bradford assay reagent, which indicates the amount of protein present by color, was added to all samples. The spectrophotometer was zeroed at 595 nm. A standard curve was made with the different absorbencies and concentrations. After the linear equation was formed, the unknown sample concentration was determined using the standard curve equation. A Gel Electrophoresis was used to perform a qualitative analysis. The use of 5 microliters of the homogenate was heated to 80 degrees Celsius. Then the homogenate was transferred to a 2-microliter-protein gel sample buffer. Samples loaded on to the gel was run at 100 v and stained with comassie blue; observations were made next lab.
