Zymography Analysis
The results from the zymography test, shown in figure 1, confirm the results in the hypotheses. In figure 1, there are two horizontal bands labeled MMP-9 and MMP-2. In addition there are twelve lanes, with the first lane representing the marker and lanes two through 12 representing the different groups in the lab. Groups 2, 4, 6, 8, 10, and 12 completed the test with PMA treated HL-60 cells and groups 3, 5, 7, 9, and 11 completed the test with DMSO treated cells. Zymography testing shows the enzymatic activity by showing varying colored bands. During the test, the MMPs eat away the gelatin just as they would eat away the extracellular matrix if they were in a living organism and as they eat away the gelatin the color begins to fade away and …show more content…
The DMSO treated cells still show some MMP-9 activity because some of these cells differentiate into monocytes, which produce MMP-9, even though most differentiate into granulocytes, which do not produce MMP-9. Monocytes are present in DMSO treated cells because DMSO is made with PMA, meaning there are trace amounts of PMA contained in DMSO. One other limitation can be seen in the differing thickness of the MMP-2 bands. This means that some of the cell samples contained more enzymatic activities, such as group 12 MMP-2 results being much thicker than the group 8 MMP-2 results. Tis could have been caused by one group loading a larger sample in than another, but this cannot be confirmed because the amount of protein cannot be seen in these results and the only thing that can be seen is the amount of enzymatic activity. Even with these limitations, the effects of differentiation on the protein can be seen because of the clear difference in MMP-9 enzymatic activity being much higher in cells treated with PMA then in cells treated with

    The homogenates provided were made by homogenizing tissues in a sucrose phosphate buffer in a 1:20 ratio. The protein concentration in bovine cells was measured by diluting the homogenate with a 1:5 ratio; 50 microliters of homogenate and 200 microliters of water. Then 5 known protein concentration samples which were 0.4, 0.8, 1.2, 1.6, 2.0 mg/ml of bovine serum were used to determine absorbance with a spectrophotometer. Two additional samples were made; one was blank and the other was for the specific homogenate sample. Then 3 microliters of bradford assay reagent, which indicates the amount of protein present by color, was added to all samples. The spectrophotometer was zeroed at 595 nm. A standard curve was made with the different absorbencies and concentrations. After the linear equation was formed, the unknown sample concentration was determined using the standard curve equation. A Gel Electrophoresis was used to perform a qualitative analysis. The use of 5 microliters of the homogenate was heated to 80 degrees Celsius. Then the homogenate was transferred to a 2-microliter-protein gel sample buffer. Samples loaded on to the gel was run at 100 v and stained with comassie blue; observations were made next lab.     For Activity A, we first tested enzyme activity. First, we used an H2O2 syringe to transfer 10 mL of H2O2 into an unlabeled 60-mL cup. Then, we used a transfer pipet to add one mL of catalase solution into the unlabeled 60-mL cup that we put H2O2 in. After that, we observed the solution for one minute. Then we tested the effect of boiling on enzyme activity. First we used a transfer pipet to transfer 4 mL of catalase into a test tube. After that, we placed the test tube filled with catalase in a boiling water bath for five minutes. While we were waiting, we rinsed the unlabeled cup we used earlier when we tested enzyme activity.
