Bacterial Growth and One Step Burst - T7 Phage
I. INTRODUCTION:
These experiments helped us learn the factors that were involved in the growth of the bacteria that increased our study towards their genetic, physical and metabolic characteristics. We used Escherichia coli and Bacteriophage T7 to identify and analyze their identical life cycle and replication that was involved in their process of growth. As, growth for any bacteria means it multiplying by increasing in its size and quantity, which can easily be examined by measuring its cell mass and cell number. Whereas, Bacteriophage T7 being double stranded makes us analyze the length of its latent period and the average burst size that occurred due to infection over a period of time. In this experiment we formed dilutions and found all the required data to analyze the process that helps in their growth, we look for colonies in Escherichia coli and formation of plaques in Bacteriophage T7. This is one way to uniquely identify them and differ them from one another.

Bacteria grown in a closed system show a specific growth pattern called the growth curve which consists of four phases. The lag phase, which is a period of slow growth; exponential phase, period of maximum growth; stationary phase, where nutrients become the limiting factor making the growth rate equal to the death rate and the death phase where organisms die faster than they are replaced. It is important to know how fast a microbe grows in order to know bactericidal or antibiotic concentrations, temperature and ph at which they stop growing. Evaluating a growth curve gives you a perspective of the generation time and the mean growth rate constant which would help you estimate the minimum, maximum and optimum growth temperature of the microbe. In this experiment you would be able to see the duration of each phase, the mean growth rate constant (k) is used to measure how fast cells are dividing in a culture, generation time, optical density (OD) which s the measure of the amount of light absorbed by a suspension of bacterial cells, and the organism’s minimum, maximum, and optimum temperatures…

This may have been due to the microscope not having a higher resolution thus affecting the accuracy of the results being obtained. Furthermore, the group MRSA also did the same practical but they used pH 2. The results that they obtained were that there were 45 cells present on the petri dish after five days. MRSA also obtained a value of 36000000 per litre. Bacteria grow by binary fission and yeast grows by budding but the patterns of growth for a population are the same.…

The population of a bacterium will grow successfully in a particular pattern known as ‘logarithmic growth’ if the location is appropriate and the proper nutrients are present. The bacteria numbers will increase and most likely continue to double in this area over time with optimal conditions. Optimal conditions meaning that there are no dangers or limiting factors, to inhibit growth in this area, such as disease, predators, competition or lack of food. At first the bacteria should reproduce slowly, this is known as the lag phase but then over time it should increase more rapidly throughout what is called the log phase. This predictable pattern of growth can be called the organism’s biotic potation.…
