Bradford Assay Lab Report
During this experiment, the method of Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE) was conducted using a negatively charged protein and pre-stained molecular weight markers. The hypothesis was that the molecular weight of N-acetyl-β-D-hexosaminidase B would be 28,000 kDa. To confirm or reject the hypothesis, the molecular weight of N-acetyl-β-D-hexosaminidase B and the concentration of protein had to be determined. The electrophoresis of the protein gel were conducted using a Hoefer SE 600 Ruby at 30 mA and stained with Coomassie Brilliant Blue Dye for 4 hours. SDS-PAGE Slab Gel Destaining Solution was then used for the removal of excess dye in the protein gel for 16 hours. In order to determine the Rf values of the protein bands, the protein gel had to be scanned using a Ricoh Super G3 scanner. Calculating the Rf values and log molecular weight values produced a standard curve with the equation of y=-1.1067x+5.2587. The antilog of the log molecular weight calculated gave a molecular weight of 79,432 kDa.

    The Lowry protein assay is a biochemical assay for determining the total level of protein in a solution. The total protein concentration is exhibited by a color change of the sample solution in proportion to protein concentration, which can then be measured using colorimetric techniques. It is named for the biochemist Oliver H. Lowry who developed the reagent in the 1940s. His 1951 paper describing the technique is the most-highly cited paper ever in the scientific literature, cited over 200,000 times. The method combines the reactions of copper ions with the peptide bonds under alkaline conditions (the Biuret test) with the oxidation of aromatic protein residues. The Lowry method is best used with protein concentrations of 0.01–1.0 mg/mL and is based on the reaction of Cu+, produced by the oxidation of peptide bonds, with Folin–Ciocalteu reagent (a mixture of phosphotungstic acid and phosphomolybdic acid in the Folin–Ciocalteu reaction). The reaction mechanism is not well understood, but involves reduction of the Folin reagent and oxidation of aromatic residues (mainly tryptophan, also tyrosine). Experiments have shown that cysteine is also reactive to the reagent. Therefore, cysteine residues in protein probably also contribute to the absorbance seen in the Lowry Assay. [3] The concentration of the reduced Folin reagent is measured by absorbance at 750 nm.[4] As a result, the total concentration of protein in the sample can be deduced from the concentration of Trp and Tyr residues that reduce the Folin reagent.The method was first proposed by Lowry in 1951.     Gel filtration chromatography is a type of column chromatography in which separated protein, peptides and amino acids on their molecular size. The stationary phase consists of beads containing pores. The mobile phase is the solvent that is found both around the beads and in the pores of the stationary phase matrix. As the sample is passes through the column, the molecule that are larger than the pores will not retarded by the beads and move only in the space between the beads. Thus, the larger molecule will not enter the stationary phase, staying in the mobile phase surrounding the beads. However, the molecule that smaller than the pores of the beads can diffuse further into the beads and move more slowly through the column. The small molecules can completely penetrate the pores in the beads are said to be included by the gel. The large molecules which can’t penetrate the pores are said to be excluded by the gel. Those that are excluded will pass through the column faster because the distance they have to travel is reduced and elute first whereas the smaller molecules will take a more convoluted path through the column and elute last.
