Purpose/Problem: There are four parts to the Enzyme Catalyst lab - Activity A, B, C, and D. In activity A, the characteristics of enzyme actions will be observed. The main purposes are to determine the rate of an enzyme catalyzed reaction, to study the characteristics of an enzyme mediated reaction, and to observe the effect of heat on enzyme activity. The purpose of activity B is to use the Titration Protocol to determine the initial amount of H2O2 present in a solution. The amount will be the baseline for activities C and D. The purpose of activity C is to determine the rate at which H2O2 spontaneously decomposes when exposed to room temperatures and ambient light for 24 hours. The purpose of activity D is to determine the rate at which catalase decomposes H2O2. After adding H2SO4 for different time lashes, etc., the resulting data will be graphed at which the catalase decomposed by catalase.…
4. Potato contains plant cells with cellulose, which is rigid and rough. The blender allows us to break through the cellulose that forms the cell wall which subsequently allows us to access the enzymes.…
In this lab the peroxidase enzyme is tested in a dormant avocado seed as well as an avocado seed undergoing the process of germination. A gas pressure will be used to test the seeds and see if the peroxidase enzyme is present in either of the seeds.…
For Figure 3, the rate of peroxidase activity greatly increased from the first to second pH, increasing .7 (AU/sec), while the second to third pH increased only .1. On the other hand, the third to final pH decreased, just like the temperature chart (Figure 2) .45 rate of activity.…
Enzymes are biological catalysts or assistants that consist of various types of proteins that work to drive the chemical reaction required for a specific action or nutrient. They can either launch a reaction or speed it up. Catalase is a common enzyme found in nearly all living organisms exposed to oxygen. It is a very important enzyme in protecting the cell from oxidative damage by reactive oxygen species (ROS). The catalase used in this experiment will come from five different sources: Spinacia oleracea (Spinach), Brassica oleracea (Broccoli), Solanum tuberosum (Russet Potato), Malus domestica (Apple), and Allium cepa (Onion). The five different catalases from the sources will all be used to catalyze Hydrogen peroxide (H2O2). When hydrogen…
Enzymes are biological catalysts (substances that speed up a chemical reaction without themselves being permanently altered)…
The enzyme peroxidase has been shown to break down H2O2. Enzymes are known to increase the rate at which a chemical reaction occurs. We looked at factors that affected the breakdown of hydrogen peroxide. These effects are the different temperatures and pH levels the enzymes were placed in. We found that the optimum, or best condition, temperature for the enzymes tested was about 22 degrees Celsius. The optimum pH level for the enzyme was 7.…
Enzymes are biological catalysts that speed up the process of chemical reactions. They are also proteins, and most enzymes activities occur within organism. They decrease activation energy, energy that is needed to start a chemical reaction. Enzymes are substrate specific substrates ending in "-ase", enzymes ending in "-ase". External factors, such as temperature, pH, and concentration of the substrate, affect the enzymes activity in the lab, pectase and cellulase will be used to compare different enzymes on apple sauce.…
Catalysts are substances that increase chemical reactions while using less energy. They also remain unchanged after use, which enables it to be used repeatedly. Cells have special catalysts called enzymes, which are specialized proteins that help accelerate chemical cell reactions. (Evert, RF & Eichhorn, SE 2013). Enzymes also control plant metabolic processes such as respiration (Evert RF, Eichhorn SE & Perry JB 2013). This experiment focuses on the enzyme catalase. Catalase breaks down hydrogen peroxide into water and oxygen. Hydrogen peroxide is a waste product of cell metabolism that can be toxic to the cell (Evert RF, Eichhorn SE & Perry JB 2013).…
Over a two week period of time in the laboratory, we experimented and tested the reaction rate of a peroxidase enzyme and the factors that affected it, both positively and negatively. The purpose of these experiments was to probe and manipulate the activity of the enzyme peroxidase by varying temperature, pH, the amount of enzyme compared to the substrate and the effect of hydroxylamine. Peroxidase activity is expressed when the potato extract is subjected to stresses such as low temperature (El-hilali et al., 2012). The most eye catching factors that we tested for their impact on enzyme activity involved change in pH, temperature, boiling extract, and the effects of probing the active site with hydroxylamine. In the first part of…
Enzymes are catalysts that speed up chemical reaction but are not themselves consumed or changed by the reaction. The cell’s biological catalysts are proteins. Proteins are made up of one or more polypeptide chains that are folded to make an active site, an area in which a material to be acted on by the enzyme, called the substrate, will fit. The temperature,pH, the concentration of enzyme, and the concentration of substrate all affect the activity of the enzyme and the rate of the reaction.…

In this laboratory exercise we studied enzyme catalase, which accelerates the breakdown of hydrogen peroxide into water and oxygen. The purpose was to isolate catalase and measure the rate of activity under different conditions. The laboratory was also conducted in association with a second laboratory that measured the effects of an inhibitor on the enzymes. Changes in temperature and pH along with Substrate Concentration and Enzyme Concentration were the conditions tested in the experiment.…
Enzyme catalysis was observed in order to analyze how changes in temperature, pH, enzyme concentration, and substrate concentration affected an enzyme-catalyzed reaction. This experiment analyzed the rate of enzyme-catalyzed reactions and observed the correlation between catalase activity and products formed. It was found out that the rate of an enzyme-catalyzed reaction starts off rapidly, decreases, and levels off or completely stops, and can be further affected by environmental factors, which play a crucial role in regulating enzymes and metabolic processes.…
Introduction: Enzymes are biological catalysts that speed up metabolic reactions without being affected. They lower the activation energy needed to start a reaction.Enzymes are affected by several factors including PH, Substrate concentration; Temperature & other factors. Each enzyme has an optimum temperature at which its activity is the highest, below this optimum temp, the kinetic energy of molecules decrease , therefore the collisions between the active site of the enzyme and substrate decreases , as a result the enzyme activity will decrease , so decreasing the rate of the reaction If the temp. Exceeds the optimum temp. The kinetic energy between molecules increase therefore collisions increase leading to the change in the tertiary structure of the enzyme and in this case active site is lost and the enzymes will be denatured so the reaction will slow down &stops. Catalase is an enzyme, found basically in all living cells. It breaks down hydrogen peroxide (waste product) into water and oxygen.…
