Principle:
To notice and examine the diabetic effect of streptozotocin which is an anti-biotic found to have a destructive effect on islets of Langerhans; beta ones which lead to disturbances in insulin production causing type 1 diabetes mellitus in animals, between different strains of rats when injected intraperitoneally.
Procedure:
• 20 adult male nude rats of average weight 250-300 mg and 8 adult male local rats of average weight 310-390 mg are kept each in a box in a temperature of 22c, supplied with food and water for 2 weeks for adaptation, during this period their weights are measured four times.

The blood was collected at T0, T1, T2 and T3..etc in a tubes then the tubes were centrifuged to collect the serum which is stored at -20C until the tests are done on it.
Oral glucose tolerance test:
To detect rats which have false low blood glucose level however they are diabetic. 13 nude rats undergo fasting for half a day (12h), then they were given a glucose solution of 2g/kg.
Blood was taken from tail before having the solution by 60 minutes and after by 30 minutes, 1 hour and 2 hours.
Results:
In the experiment 20 nude rats and 8 local rats were used, before induction of diabetes by streptozotocin the blood glucose of nudes' ranges between 119 mg/dl-146 mg/dl and in local rats ranges between 120 mg/dl- 164 mg/dl. After injection in nudes the blood glucose increased by a percentage of 0.05 while in local rats their blood glucose become above 300 mg/dl.
The mean of levels of glucose in local rats ranges 339 mg/dl – 490 mg/dl and that happens in only 7 of them while the rest ranges from 167 mg/dl – 275 …show more content…
Procedure
• The procedure of injecting the rabbit was held in a steel holder designed for blood collection.
• They are injected intravenous to the marginal ear veins using insulin needle on 10 syringe or less if required dabbing of xylene to particular vein.
• The xylene would make the ear veins prominent and easy to inject to determine random blood sugar level.
Discussion
When the alloxan administrated on the animal it is concentrated in the islets of Langerhans and liver to be reduced into dial uric acid which is unstable then oxidation will happen it will back to alloxan and generation of O2 and OH radical will happen. It was discovered that alloxan to have more effective action must be selective cytotoxic to beta cells of pancreatic islets of Langerhans. Alloxan also produce antioxidant enzyme superoxide activity in islet cells.
Results
It is resulted that the administration of alloxan has high increase in the blood sugar level of the rabbits. It also resulted that alloxan should not be administrated by single dose 160 kg so they should give orally glucose per kg before administrating
