Kristen Magliola 9/9/13 BIO: 414L Lab Report: SOC Medium

I. Purpose: To make liquid and solid media for microbial growth

II. Background: There are four different types of media used to grow microbes. They consist of Enriched media, Nutrient media, Selective media, and differential media.
Enriched media is a growth media supplemented with complex biological molecules. This type of media is used for blood, coagulated blood, amino acids, etc. Nutrient media provides nutrients necessary for the growth of microorganisms. Selective media is used for specific organisms. For i.e. Mackonkey agar only grows on gran(-). If a sample is negative it grows and if it is positive it doesn’t grow. Last but not least Differential media relies on color change of the media itself. This color change highlights the bacteria if it has a specific trait. For i.e. If sugar ferments there will be a color change.

III. Reagents:
1.Tryptone
2.Yeast extract
3.NaCl
4.KCl
5.MgCl2
6. MgSO4
7.Glucose/ Dextrose
8.H20
9.Agar( for solid)

IV. Methods
Grab an Erlenhymer flask for the solid solution and another flask for the liquid solution. Label both flasks.
Put 200 ml of distilled water in each flask
Then weigh out each of the reagents.
Pour the reagents into each flask
Swirl/mix the liquid flask. When everything is dissolved cap it off
Add 8g of agar to the Erlenhymer flask
Take out the Bunsen burner, turn it on, and put the flask on the burner
Check the flask every now and then and swirl (for approx. 5 min) The media is done when the particles are dissolved

VI. Results/Conclusion: The purpose of this lab was to make liquid and solid media for microbial growth. The pictures above are evidence of the media. The media that was supposed to turn solid did just that( as seen on the left). Solidified agar indicates success. Another step was performed after the original liquid media was made. The liquid media was autoclaved. The autoclave uses heat and

    In this lab experiment we did several test to determine what our unknown bacteria was. To determine this we recorded the results of how the bacteria reacted to different media. Depending on the results of each test we could narrow down the different bacteria to determine what our unknown is. This experiment will also determine if our bacteria is a fermenter of sugars and if it is catalase positive. If the bacteria is a fermenter they will use the sugars to make ATP.
