Tacrolimus Research Paper
The major elimination process of Tacrolimus metabolites include biliary route, about 95 % of elimination takes place here (Buijsch, 2007). Urinary excretion accounts for about 2.4 % Tacrolimus elimination. Concentration of Tacrolimus metabolites is reported to increase through the biliary distraction (Buijsch, 2007). Tacrolimus metabolism goes through o-demethylation, hydroxylation and oxidtie metabolic reaction by the enzymes of , CYP3A4 and CYP3A5, in the liver and the intestinal walls. Cytochrome P450 , 13-O-demethyltacrolimus, demethylhydroxy tacrolimus, and hydroxy tacrolimus are some of the few metabolites produced by the the two step reaction (Gonschior, 1996). These metabolites contribute to the 42% blood concentration in the liver and 44.8% in the kidney (Gonschior, 1996). The blood concentration for the therapeutic range is higher in the kidney than in the liver, because the activity of the Cytochrome P450 enzymes is lower in the liver than it is in the kidney (Gonschior, 1996). …show more content…

Adult male rats received a single subcutaneous dose of 0.1 mg per kg dulaglutide, and blood samples were collected 1,2,4 and 6 days following administration. A group of 3 monkeys received a subcutaneous dose of 0.1 mg per kg dulaglutide and blood samples were collected at 0, 2, 4, 12, 48, 72 96, 192, 240, 288 and 336 hours following administration. Samples from both animals were studies for GLP-1-Fc concentration using ELISA, utilizing antibodies that recognize the N-terminus of GLP-1-Fc and the Fc domain. Optical density of tetramethylbenzidine development was measured and concentrations of GLP-1-Fc were calculated using the four parameter algorithm (Glaesner, et al., 2010). The pharmacokinetic parameters of dulaglutide for rats and monkeys respectively were: t1/2= 38.2 hours, and 51.6 hours; Cmax = 179.7 ng/mL, and 292.2 ng/mL; Tmax= 24 hours, and 16.7 hours; Cl = 9.6 ml/h/kg, and 7.3 mL/h/kg; VD = 525.0 mL/kg, and 557.5 mL/kg; AUC0- ∞ = 10,537 and 15,207 (Glaesner, et al., 2010; Jimenez-Solem et al.,…

    Clopidogrel Case Study

Clopidogrel is a thienopyridine prodrug that requires hepatic biotransformation to form an active metabolite that selectively and irreversibly inhibits the purinergic P2RY12 receptor, and thereby platelet aggregation, for the platelet's life span (~10 days). Only 15% of the prodrug is available for transformation to the active agent; the other 85% is hydrolyzed by esterases to inactive forms. Conversion of clopidogrel to its active metabolite requires two sequential oxidative steps involving several CYP450 enzymes (e.g., CYP1A2, CYP2B6, CYP2C9, CYP2C19, and…
