Enzyme Peroxidase Lab Report
Enzymes are important components of life, facilitating reactions that are necessary for an organism to live. Enzymes can be very specific to what environment they function best in1. Numerous environmental impacts were tested for the enzyme peroxidase which catalyzes the decomposition of hydrogen peroxide. The basic decomposition reaction was carried out first without any environmental alterations. The hypothesis for this reaction was supported. The enzyme caused the amount of absorbance increase as time went on at a fairly constant rate (Figure 1). As shown in Figures 2A and 2B, pH has a large effect on the rate to which an enzyme catalyzes a reaction. The optimum pH for the peroxidase was 6. Each varying pH produced a different reaction rate. The pH of 3 and the pH of 9 caused the peroxidase to work at its slowest. The enzyme worked best at pH 6 which was closely followed by pH 7. Peroxidase depends on specific pH’s to be effective, most likely based upon the environment it is usually found in. The hypothesis that the different pH values would have varying effects on the rate to which peroxidase catalyzes the reaction was supported. Altering the enzyme concentration also had a great impact on the rate of the decomposition of hydrogen peroxide. As the concentration of the enzyme increased, so, too, did the rate of the reaction. The reaction was tested with 0.1 mL of H2O2, 0.2 mL of H2O2, and 0.4 mL of H2O2. All three rates of reactions calculated were within 0.0007 which is a very miniscule difference. The rates of reaction could be very similar since the amount of enzyme was not increased, so there was the same number of enzyme molecules reacting at their optimum pace while more and more substrate molecules were added.     This experiment was performed to determine the resultant effect of temperature change on the reaction between the enzyme catalase and hydrogen peroxide. This experiment was performed by measuring and comparing the amount of oxygen bubbles produced and the absorbance of the catalase and hydrogen peroxide solution over time at room temperature, 2°C, 50°C, and 60°C. The overall result of this experiment proved that this reaction works best at room temperature, slows down when the temperature is lowered, and does not take place when the temperature gets too high due to the enzyme denaturing at these higher temperatures. The hypothesis that was expected for this experiment was supported by the actual experiment for the most part, but due to an error in the absorbance measurement, the results were somewhat skewed leaving part of the hypothesis unproved for this part of the experiment.     Enzymes are catalysts to ensure homeostasis in the body [2]. Homeostasis is the tendency to maintain the internal environment [2]. Hydrogen peroxide is an oxidizing agent that can be decomposed by catalase [11], the liver is able to decompose hydrogen peroxide enzymatically because it contains catalase [12]. The reaction rate of an enzyme catalysed reaction under the effect of changes in temperature was tested in this experiment. It was hypothesised that the fastest reaction rate would occur a 37 degrees Celsius. Liver and hydrogen peroxide were placed into 4 different test tubes, all set in different temperatures: 0 degrees Celsius; room temperature (28 – 30 degrees Celsius); 37 degrees Celsius; and 100 degrees Celsius. The results occurred as predicted, on graph they formed a semi-circle like shape. The 37 degrees Celsius delivered the best result with the room heat giving a close comparison but the iced and boiling heat delivered minimal results.     Our results indicate that the presence of iron increases the rate of the peroxidase’s activity, judging from the increase in absorbance with increasing amount of iron. This result supports our hypothesis that the iron acts as a cofactor in peroxidase’s activity and results in the faster rate of the reaction. Also, we assumed that the more we put iron in the solutions, the faster the rate is. This assumption is also supported by the results that higher concentration of iron cofactors shows the higher absorbance. The importance of the existence of iron in this reaction is that the peroxidases in plant cells respond to environmental stress by, in this case, catalyzing the conversion of toxic metabolic wastes, H2O2, into the harmless products, H2O and O2, faster. Therefore, as peroxidases help minimize and eliminate harmful chemicals, the iron cofactors that help peroxidase’s activity raise the possibility of plants’ survival in harsh environments.…

Our second lab, we tested enzyme and substrate concentrations to determine the increase of temperature and inhibitor. The enzyme source used in both labs was peroxide, guaiacol is used as a substrate for peroxide. We used Guaiacol, turnip extract, peroxide and distilled water for enzyme and substrate concentration. In the second lab we used the same substances in lab one, however using the temperatures of 4○C (ice bath) , 23○C ( room temperature), 37○C (body temperature) and 60○C (water bath) to determine increase of temperature.     Conclusion: This experiment was to test if increased amount of pH will affect the rate of reaction in an enzyme catalyzed reaction. The hypothesis was correct in stating that the increased pH will increase the rate of reaction. In Test one the qualitative color was only at nine after five minutes of timing the reaction. In the next experiment the qualitative color was at ten, for some, before the four minute mark. This being the case, the experiment does not support our hypothesis. The graph above shows the rate of reaction. Within the first three minutes of the reaction drops at a very rapid rate. Then, the next two minutes the rate of reaction has a huge increase. After this wave the reaction steadily decreases until it halts. Since, at the beginning of the reaction the reaction dropped, the color changed very slowly. With the increase the color changed very dramatically. In conclusion, increasing amount pH to the reaction did help increase the time it took to finish.…
