Separating Water Soluble Ink Through Chromatography
Separating Water Soluble and Acetone Soluble Inks through Chromatography

Abstract:

Chromatography is a term used to define a set of laboratory techniques for the separation of mixtures. Through Chromatography we analyzed complex mixtures. In our recent lab we analyzed complex mixtures. The purpose of this lab was to find whether the water soluble or acetone soluble liquid traveled farther. In our recent lab we measured the distance traveled by the different colored water and acetone soluble inks. W put three dots of water soluble ink on a piece of filter paper and four dots of acetone soluble ink at the other end. We then put the end of the filter paper in a small amount of distilled water in a beaker. The filter paper absorbed the water and the ink slowly spread to the top of the paper. We repeated these steps for both the water soluble and acetone soluble ink but when we did the acetone soluble ink, we placed the filter paper in an acetone-water mixture instead of just distilled water. We then recorded our results which showed that water soluble inks traveled farther than the acetone soluble inks. The results of our experiment showed how each different color of water soluble inks traveled farther than any of the acetone soluble inks.

On the lines drawn we put four circles filled with water soluble ink. When the ink dried, we put a second drop of ink in each circle. We then put the filter paper in a 400 mL beaker filled with ¼ inch if distilled water. (The filter paper was folded length wise.) The beaker was then covered with plastic paper wrap and we watched the dots spread to the top of the paper as it absorbed the water. We then took the paper out quickly and measured the distance the ink traveled. We then repeated these steps with the acetone soluble ink except ¼ inch of acetone instead of distilled

The first step was to prepare the two filter paper for the experiment so you can measure the different columns at the end of the experiment. For that to happen you have to draw a horizontal line with a pencil. The line has to be 1cm above the bottom. After that you draw nine cross lines along the horizontal line with a space between each cross line of 1.5cm. Now you have to mark each area with a shortcut of the substance you are going to put on the horizontal line. The next step is to prepare each of your substances in the 24-well plate. Once you put all the substances in the well plate and dissolved the color from the M&Ms and Kool Aid with some drops of water, you dip a clean toothpick in the dye. You mark carefully a little point on the horizontal in the center of the section you marked with the substance and make sure the point is not too big because otherwise the experiment won’t work as good. The filter paper needs to dry for a few minutes before you form both to a cylinder and staple it so it stays a cylinder. Before you can keep going with the experiment you have to take a 50ml plastic beaker and fill that one with 50ml distilled water, solve a pinch salt in the water and stir until you can’t see crystals anymore. Pour a little bit of that solution in a petri dish but make sure it will be below the horizontal line you draw on the filter paper. Set…

In order to experiment with distinct components of a mixture, they must be first separated so they can be observed individually. This is accomplished in this lab by the technique called thin layer chromatography (TLC). TLC involves a stationary phase, which the TLC plates as well as a mobile phase, which could be one of two solvents used: ethanol-acetone for TLC. Dyes in a sample separate consequently because of their unique polarities. As a result, nonpolar substances travel further than polar substances in this process. The separation of a mixture into its components by TLC transpires because the distinctive components of a mixture interact to different extents with the stationary and mobile phases.…

Chromatography serves mainly as a tool for the examination and separation of mixtures of chemical substances. Chromatography is using a flow of solvent or gas to cause the components of a mixture to migrate differently from a narrow starting point in a specific medium, in the case of this experiment, filter paper. It is used for the purification and isolation of various substances. A chromatographically pure substance is the result of the separation. Because purification of substances is required to determine their properties, chromatography is an indispensable tool in the sciences concerned with chemical substances and their reactions.…

The chromotography project was left open to variation and interpretation from team to team. Initially, my team struggled with the many choices for markers and filter paper and our choices may have affected our results. For example, we had the choice between a coffee filter and a circular piece of filter paper. We originally chose the coffee filter but we thought the creases in our paper would distort our results. After finally deciding on the circular paper and cutting it into a rectangle, I placed it in the beaker to see if it fit properly. Unfortunately, I did not notice that the beaker had water in it and had soaked the edge of the filter paper. Although I do not know for sure, this may have altered our results. Since a piece of the paper…

Candy Chromatography is the method of analysis of separating and comparing dyes used in candies and food coloring. It is also a proven method to analyze candy and its complex structure, without having to taste it. These complex mixtures can be separated back into pure substances. To find out the ingredients of a favorite candy, chromatography would help to define every ingredient found in the candy sample. The word chromatography is derived from the two Greek words, “Chroma” meaning color and “graphic” meaning to write. Paper chromatography is the best method of separation used in candy chromatography. Paper chromatography is a technique that involves placing a small dot or line of sample solution onto a strip of chromatography paper.. The paper is placed in a jar containing a shallow layer of solvent and sealed. As the solvent rises through the paper, it meets the sample mixture, which starts to travel up the paper with the solvent. This paper is made of cellulose, a polar substance, and the compounds within the mixture travel farther if they are non-polar. More polar substances bond with the cellulose paper more quickly, and therefore do not travel as far.…

Pigments extracted from different greens have different polarities and may be different colors. Mixed pigments can be separated using chromatography paper. Chromatography paper is able to separate mixed pigments due to their polarity and solubility. Pigments of chlorophyll a, chlorophyll b and beta carotene will be separated on chromatography paper because each has its own polarity and solubility, which results in different distance traveled up the paper. Beta carotene is non-polar so it travels the highest distance, followed by chlorophyll a. Chlorophyll b is the most polar; therefore, it travels the shortest distance. The separated pigments on the chromatography paper can be eluted in acetone and absorbance spectrum is determined using spectrophotometer. Spectrophotometer produces a graph of the absorbance spectrum which shows the wavelengths (in nm) that are absorbed by the pigment and that are reflected. The reflected wavelength of chlorophylls a and b reflect the wavelengths of l light that are associated with green, which gives them their color. Beta carotene reflects the wavelengths that are associated with orange color, and that explains its color.…

Chromatograph method is a method of separating mixtures of two or more compounds. Two phases are important in this method; one that is stationary and one that is moving. Chromatography works on the principle that different compounds will have different solubilities and adsorption to the two different phases between which are to be partitioned. Thin Layer Chromatography (TLC) is a solid-liquid technique. The mixture is observed when it is in two different phase; a solid (stationary phase) and a liquid (moving phase). Thin Layer Chromatography (TLC) is a technique which is sensitive, cheap and fast. A thin layer chromatograph plate is used to identify drug component under UV. It is used to support the identity of a compound in a mixture when the Rf of a compound is compared with the Rf of a known compound.…

Chromatography can be described as a mass transfer process involving adsorption. HPLC relies on pumps to pass a pressurized liquid and a sample mixture through a column filled with a sorbent, leading to the separation of the sample components. The active component of the column, the sorbent, is typically a granular material made of solid particles (e.g.silica, polymers, etc.), 2–50 micrometers in size. The components of the sample mixture are separated from each other due to their different degrees of interaction with the sorbent particles. The pressurized liquid is typically a mixture of solvents (e.g. water, acetonitrile and/or methanol) and is referred to as a "mobile phase". Its composition and temperature plays a major role in the separation process by influencing the interactions taking place between sample components and…
