Comparative Analysis Of Unknown Bacteria
The main purpose of this experiment is to identify the unknown bacterium in a mixed culture using comparative analysis of morphological and biochemical characteristics. Using aseptic technique, the isolated unknown bacterium was inoculated in different types of media and incubated for 24 hours and then stored in 4° C. Different types of bacteria show different results in these tests, so some specific tests were done to narrow down the possibilities. After performing various biochemical tests such as H2S, Indole, Motility, MR, VP, Citrate, Glucose, Lactose, Mannitol and Catalase tests, the given unknown bacteria 02-09 was found out to be Pseudomonas

The Unknown Gram Negative bacterium inoculated in a Tryptic Soy broth medium was randomly selected from a group of other unknowns. In order to identify this unknown the seven different types of biochemical tests will be conducted on this unknown bacterium to identify it out of 6 possible bacteria; Escherichia coli(E. coli), Enterobacter aerogenes (E.aerogenes), Klebsiella pneumoniae (K.pneumoniae), Proteus mirabilis(P.miranilis), Pseudomonas aeruginosa(P.aeruginosa), and Salmonella typhimurium (S.typhimurium). The biochemical tests utilized were; Triple Sugar Iron Agar (TSIA) test, Sulfur Indole Motility (SIM) test, Methyl red test and Voges-Proskauer (MR-VP) test, Citrate test, Urea test,…

The purpose of this experiment was to isolate two unknown bacteria and perform a series of selective and differential tests to correctly identify each. After the bacteria was isolated a series of differential and selective tests following the dichotomous key attached were used to identify each bacteria. The Gram-positive bacteria were identified as Staphylococcus aureus with a positive confirmatory test, mannitol salt agar, showing consistent results as well for S. aureus. The Gram-negative bacteria were Pseudomonas aeruginosa with a positive confirmatory…

The purpose of the unknown bacteria lab assignment was to select an unknown bacteria culture and, through a series of metabolic tests, identify which bacteria genus resided in the pure culture received. A nutrient broth inoculated with bacterial culture (numbered 45, henceforth referenced as U45) was selected and a streak plate was made to isolate a pure culture for use throughout the assignment.…

In this lab experiment we did several test to determine what our unknown bacteria was. To determine this we recorded the results of how the bacteria reacted to different media. Depending on the results of each test we could narrow down the different bacteria to determine what our unknown is. This experiment will also determine if our bacteria is a fermenter of sugars and if it is catalase positive. If the bacteria is a fermenter they will use the sugars to make ATP. If the bacteria is a fermenter of lactose/sucrose the EMB plate we used will “clearly differentiate between the colonies of lactose fermenting and non-fermenting microbes. In the same medium sucrose was also included to differentiate between coliforms that were able to ferment sucrose more rapidly than those that were unable to ferment sucrose” (Cheeptham & Lal, 2007).…

In the unknown identification labs, we have identified our unknown as Pseudomonas aeruginosa. Pseudomonas aeruginosa is Gram negative and rod shaped that we found to be motile in the lab. Our strain of P. aeruginosa formed colonies that were round in shape and had scalloped margins on nutrient agar. On our agar slant, the P. aeruginosa colonies had a filiform appearance on the edges.…

The purpose of this lab was to identify two unknown bacteria cultures using various differential tests. The identification of these unknown cultures was accomplished by separating and differentiating possible bacteria based on specific biochemical characteristics. Whether the tests performed identified specific enzymatic reactions or metabolic pathways, each was used in a way to help recognize those specifics and identify the unknown cultures. The differential tests used to identify the unknown cultures were oxidase, catalase, lactose and sucrose fermentation, Kugler/iron agar, nitrate reduction, gelatin hydrolysis, starch hydrolysis, manitol salt, MR-VP, citrate, bile esculin, indole, urease, DNase, and coagulase.…

An unknown bacterium experiment, this experiment was done to discover what bacterium was in a mixed culture sample. This experiment worked with enteric bacteria which are members of the Enterobacteriaceae and live in the intestinal tract (Willey et al., 2014). Also found in Willey et al (2014) is some characteristics of enterics such as they all degrade sugars by means of the Embden-Meyerhof pathway, they produce plasmid-encoded proteins and that some of them perform mixed acid fermentation, some use enzyme systems like formate dehydrogenase and pyruvate formate-lyase (PFL) The first thing that had to be done was the streak plating to isolate the bacterium, then the next lab multiple tests were ran to help identify which bacterium was in the sample. One of the most important tests done was gram staining because it helped to limit the possible bacterium that it could have been. To be sure which bacterium was worked with we referred to the Bergey’s manual and compared the results to the possible bacterium.…

The plate count agar, CFC agar and cetrimide agar are used and provided different evidences on the isolation of Pseudomonas from the soil experiment. Firstly, the plate count agar is a medium for the enumeration of viable organisms in food, water, waste water and also from clinical samples, and thus it is non-selective to any species. Whereas the CFC agar is a selective agar which contains reduced amounts of cetrimide but also cephaloridine and fucidin are added to allow the isolation of most Pseudomonas species. If the Pseudomonas species are present, a blue-green or brown pigmentation under the daylight, or fluorescence can be taken as a presumptive evidence under UV light. In the experiment, we had observed the colonies under UV light and…

The unknown microorganism #5 is Pseudomonas aeruginosa. It can survive in a variety of environments and at temperatures found in clinical settings (Murray et al, 1999). Clinical setting is referring to temperatures such as those in the hospital environment. P. aeruginosa can also survive in aqueous environments. Pseudomonas aeruginosa is found at several different sources. Nosocomial sources of this microorganism are: respiratory therapy equipment, eye drops, dialysis fluid, and disinfectants. It can also be found in the GI tract and moist areas on the human body. Some other sources of Pseudomonas aeruginosa are: contact lens solution, cosmetics, and pools. However it is not commonly found as the normal flora on the human…

This lab focuses on the isolation and characterization of an unknown organisms expressing interesting properties in relation to Antimicrobial Activity. At the conclusion of this laboratory experience The researcher would be required to describe the isolates obtained from the soil macroscopic and microscopic appearance, Perform the gram stain of the microorganisms in question and to discriminate the organisms from other microbes that could be contained in the soil and to finally make certain that the organisms have not been exposed to outside sources and be isolated in a pure culture. The researcher must first isolate the microbe and try to grow the organism in a pure culture to commence with testing the organism. The isolation of Microbes in a laboratory and clinical settings are of the up most importance and due to the isolation of different microbes from each other various procedures become open for the…
